植物生长延缓剂多效唑和比久对怀山药离体保存的影响

摘  要:
为了寻找怀山药种质资源缓慢生长离体保存的有效方法,以铁棍山药、47号山药和B号山药为试材,研究了植物生长延缓剂多效唑(PP333)和比久(B9)对其试管苗在常温(25℃)和低温(4℃)下的保存效果。结果表明:常温和低温下,PP333、B9均可延缓各怀山药试管苗的生长,PP333的保存效果优于B9的,低温的保存效果优于常温的。在MS+KT 2mg/L+NAA 0.02mg/L培养基上,铁棍山药、47号山药和B号山药低温保存的适宜PP333质量浓度分别为5.0mg/L、2.5mg/L、1.0mg/L,在此条件下,各怀山药无需继代可保存6个月,保存6个月的存活率分别为100%、100%、95%。在试验范围内,怀山药种质资源缓慢生长离体保存的有效方法为:将不同怀山药带芽节段接种在添加各自适宜质量浓度PP333的MS+KT 2mg/L+NAA 0.02mg/L培养基上,常温下培养7d,再转入4℃低温、自然光下保存。

摘  要:
In order to find the effective method for retarding growth of Dioscorea opposita Thunb.germplasm resources in vitro,in this study,effects of PP333 and B9 on preservation of D.opp.Thunb.test-tube plantlets were studied using D.opp.Thunb.cv.Tiegun,47 and B as experimental materials.The results showed that at room temperature(25 ℃)or low temperature(4 ℃),PP333 and B9 all could retard growth of Tiegun,47 and B test-tube plantlets,the preservation efficiency of PP333 was better than that of B9,and the preservation efficiency of low temperature was better than that of room temperature.On the medium of MS+KT 2 mg/L+NAA 0.02 mg/L under low temperature,the optimum mass concentration of PP333 for preservation of Tiegun,47 and B plantlets was 5.0 mg/L,2.5 mg/L and 1.0 mg/L,respectively.Under the above condition,the test-tube plantlets could be preserved for 180 days without subculture;their survival rates were 100%,100% and 95%,respectively.In a word,the best method for retarding growth of D.opp.Thunb.germplasm resources in vitro was as follows: test-tube plantlets were cultured for7 d on the MS+KT 2 mg/L+NAA 0.02 mg/L added with the optimum PP333 at room temperature,and then transferred to low temperature and preserved under natural light.
