水稻花青素合成调控转录因子OsC1和OsPAC1的功能

摘  要:
从黑米品种黑帅中克隆水稻内源调控花青素合成的转录因子OsC1和OsPAC1,并导入到白米粳稻品种Chao 2-10和ZH11中超量表达,结果显示:在超量表达OsC1的Chao 2-10的抗性愈伤分化阶段、再生转基因植株的叶片、叶鞘、稃尖等部位观察到花青素的积累;其他转基因材料,如超量表达OsC1的ZH11、超量表达OsPAC1的Chao 2-10和ZH11均未观察到花青素的积累;利用高效液相色谱(HPLC)测定转基因水稻叶片中花青素含量,发现主要是矢车菊素-3-O-葡萄糖苷和芍药素-3-O-葡萄糖苷2种花青素。对转基因材料中花青素生物合成相关基因的表达量进行分析发现,Chao 2-10和ZH11超量表达OsC1后,花青素代谢途径上的大部分结构基因上调表达;Chao 2-10中超量表达OsPAC1后,OsCHS、OsCHI、OsANS、OsUFGT上调表达,而OsF3 H、OsF3’H表达基本没变;ZH11中超量表达OsPAC1后,OsCHS、OsANS、OsUFGT上调表达,而OsCHI、OsF3 H、OsF3’H的表达基本不变。

摘  要:
Rice endogenous transcription factor OsC1 and OsPAC1,which may regulate the biosynthesis of anthocyanidin,were isolated from black rice Heishuai and overexpressed in white rice Chao 2-10 and ZH11.Anthocyanin accumulation was observed in the resistant callus,leaves,leaf sheath,apiculus of OsC1-overexpressed Chao 2-10.Results of high performance liquid chromatography(HPLC)analysis showed that anthocyanins cyanidin-3-O-glucoside and peonidin-3-O-glucoside were accumualted in OsC1-overexpressed Chao 2-10.No anthocyanin accumulation were observed in other transgenic rice materials including OsC1-overexpressed Chao 2-10,OsC1-overexpressed ZH11 and OsPAC1-overexpressed ZH11.Expression analysis of structural genes related to the pathway of anthocyanin biosynthesis in transgenic rice showed that most of the structural genes involved in anthocyanin biosynthesis were up-regulated in OsC1-overexpressed Chao 2-10 and ZH11except OsDFR.Expressions of OsCHS,OsCHI,OsANS and OsUFGT were up-regulated in OsPAC1-overexpressed Chao 2-10,but the expression levels of OsF3 H and OsF3′H were not changed.Expressions of OsCHS,OsANS and OsUFGT were up-regulated in OsPAC1-overexpressed ZH11.The expression level of OsCHI,OsF3 H,and OsF3′H kept unchanged.
