大豆花芽差异表达基因片段GmFBDG01克隆和RNAi表达载体的构建

摘  要:
大豆的产量构成因素中每荚粒数是提高大豆产量的育种关键因素,因此研究与荚粒数相关的基因至关重要,花芽作为植物生殖生长的第一步,对大豆植株的生长发育有着重要的影响,可能对四粒荚的产生有影响。以大豆吉农18四粒荚突变体的7叶期花芽的转录组测序结果进行分析,筛选出差异表达的片段Gm FBDG01;利用RT-PCR克隆差异表达基因的CDS核心序列,并借助中间克隆载体p Bluescript SK plus,成功构建了RNA干扰(RNAi)表达载体p CPB-Gm FBDG01-RNAi,该表达载体的构建为大豆四粒荚相关功能基因的功能验证奠定基础。

摘  要:
As the first step of reproductive growth of plants,flower bud has an important influence on soybean plant growth and development. Choosing flowering buds in the stage of the seventh compound leaf in soybean four-grain pod mutants of jinong 18 as materials,this study analyzed and compared results of the trans criptome sequencing. Gm FBDG01 core sequence of differential expression gene was cloned by RT- PCR,and soybean RNA interference expression vectors p CPB-Gm FBDG01-RNAi were successfully constructed with intermediate cloning vector p Bluescript SK plus. The results will lay foundation for functional verification of related functional genes of soybean four-grain pods.
