栽培种花生ddGBS测序中酶切组合的选择

摘  要:
花生全基因组水平生物信息的挖掘是研究花生种质遗传资源的前提。基于已公布的两个栽培种花生的祖先二倍体(Arachis duranensis和A.ipaensis)的全基因组序列,根据双酶切GBS(Double digest Genotyping-by Sequencing,ddGBS)测序特点,通过SacⅠ和Mse Ⅰ、PstⅠ和Msp Ⅰ、EcoR Ⅰ和NIaⅢ的电子酶切结果,统计和对比三组酶切片段的均匀度和覆盖度,筛选得到ddGBS测序片段所需的最佳酶切组合EcoR Ⅰ和NIaⅢ,并进一步揭示了其酶切片段在染色体水平及全基因组水平的分布情况,确保该酶切组合在后续基因组信息挖掘中的可行性,为全基因组层水平上遗传资源的揭示奠定基础。

摘  要:
The whole genome sequencing is the foundation for studying the genetic resources of peanut.The genome sequences of two ancestral diploid peanut have been published and therefore provided important references of this cultivated tetraploid.This study will aim at screening the available restriction enzyme combinations of ddGBS(Double digest Genotyping-by-Sequencing).The electronic restriction enzyme method was adopted to evaluate the fragment distributions of three restriction enzyme combinations(Sac Ⅰ and Mse Ⅰ;Pst Ⅰ and MspⅠ;EcoRⅠ and NIa Ⅲ).The restriction enzyme combination,EcoRⅠ and NIa Ⅲ,generated well-distributed sites,which will provide the solid step for genetic resource study at the whole genome level.
