绵羊外周血中游离DNA提取及SRY基因检测

摘  要:
【目的】外周血中游离的DNA主要是指存在于血浆/血清等循环系统里的一些小片段DNA,应用在一些疾病的诊断以及非创伤性产前诊断方面。研究三种方法从绵羊外周血浆/血清中分离提取游离DNA,建立高效的外周血游离DNA提取方法,并对SRY基因进行PCR扩增。【方法】利用加热法、酚氯仿法以及试剂盒法对500μL血浆/血清中游离的DNA进行提取。根据游离DNA片段大小的特点分别设计扩增产物大小为168 bp的检测引物和178 bp的SRY基因引物进行普通PCR检测。【结果】利用加热法、酚氯仿法、试剂盒法都提取到了游离的DNA,PCR后通过琼脂糖凝胶电泳检测到了168 bp的目的片段。同时利用PCR扩增到了178 bp的SRY基因目的片段。【结论】不同的提取方法均可有效提取分离绵羊外周血血清和血浆中游离的DNA片段,能够用于SRY基因的检测。

摘  要:
【Objective】Free DNA of peripheral blood mainly refers to some small fragment DNA which exists in plasma and serum.It has a great application prospects in the diagnosis of some diseases and non invasive prenatal diagnosis.【Method】 The purpose of this study is to establish an efticient free DNA extraction protocol from peripheral blood of sheep by three methods.Then the SRY gene is likely to be detected by PCR amplification.The free DNA of plasma and serum was extracted with the heating method,phenol chloroform method and kit.According to the characteristics of the free DNA,we designed suitable primers,whose amplification produet size was 168 bp and 178 bp respectively.【Result】 The free DNA were extracted by three methods and there were 168 bp band detected by garose gel electrophores after PCR.At the same time the SRY gene was detected to be 178 bp band.【Conclusion】 Different extraction methods are effective to extract free DNA in peripheral blood serum and plasma of sheep,which can be used for the detection of SRY gene.
