荧光抗体诊断水牛巴贝斯虫病的研究

摘  要:
用间接荧光抗体试验(IFAT)和直接荧光抗体试验(FAT)对水牛巴贝斯虫病进行了研究。IFAT结果表明,我国的水牛牛巴贝斯虫和澳大利亚黄牛牛巴贝斯虫与伊氏锥虫、环形泰勒虫、瑟氏泰勒虫相边缘边虫无交叉免疫反应,而与双芽巴贝斯虫有较低的交叉免疫反应。IFAT和FAT交叉免疫反应及FAT抑制反应证明,国内水牛牛巴贝斯虫、黄牛牛巴贝斯虫和澳大利亚黄牛牛巴贝斯虫三者抗原性相同。用IFAT测定,实验水牛感染后3—8天产生抗牛巴贝斯虫抗体,第23天达到最高峰(1:10240),并维持在一个较高水平。感染后第143天抗体效价仍为1:1280,而水牛血液中的虫体已于感染后第23天消失。水牛巴贝斯虫病疫区内的水牛和黄牛血清学调查显示,100%抗体阳性(水牛16/16,黄牛12/12),而非疫区水中则100%抗体阴性(29/29)。水牛牛巴贝斯虫玻片抗原在4℃.—30℃相—70℃下分別深存3.5个月、1年和1年,其抗原性无明显变化。荧光抗体技术具有很高的敏感性和特异性,是一种有效的诊断水牛巴贝斯虫病的方法。

摘  要:
The authors described the indirect fluorescent antibody test(IFAT)and fluorescent antibody test (FAT) for detection and studies of Babesia bovis antibody in buflalo serum and B. bovis. The results showed that there was no cross-reaction between B. bovis and anyone of Trypcnosoma evansi, Theileria sergenii, T. annulata and Anaplasma Marginale, but a lower cross reaction between B.bovis and B.bigemina,IFAT and FAT results showed that two strains of B.bovis in buffalo and cattle from China and one strain of B. bovis in cattle from Australia belong to the same species, B.bovis.Antibody against B. bovis measured by indirect fluorescent antibody test in experimentally infected buffaloes were positive at 3-8 days after infection, reaching a peak at 23 days after infection,with maximum titre up to 1:10240,and maintaining at a high level, till 143 days after infection. However the parasites became undetectable by the 22nd day post infect ion.The serological surveys of B.bovis of buffaloes and cattle were carried using IFAX. In B. bovis IFAT,antibodies of the sera from enidemical areas were all positive, but those of the sera of buffaloes from non-epidemical areas were all negative.The infected blood smear antigens were preserved at 4℃, -30℃ and -70℃ for3.5 months, and one year respectively, but their antigenicities were not changedappreciably.IFAT and FAT are sensitive and specific methods to diagnose and study theinfection of B.bovis.The results of the immuno-cross-reection tests suggested a homology of buffaloand bovine serum protein, particularly 1gG.
