摘  要:
为了对8个尼罗罗非鱼家系进行亲子鉴定,利用GM024、GM435、GM459、UNH719、UNH846、GM558六个微卫星标记设计引物并进行PCR扩增,在12%聚丙烯酰胺凝胶上电泳分型,应用cervus3.0软件统计分析。结果表明,各微卫星位点的等位基因数在4~12之间;观测杂合度在0.313~0.988之间,平均观测杂合度为0.696;期望杂合度在0.318~0.794之间,平均期望杂合度为0.638。平均多态信息含量为0.608。已知一亲本时,各个位点的非父排除率在0.301~0.818之间;双亲未知时,各位点的累计排除率在0.113~0.69之间。当已知一亲本时,6个位点的累积排除率达到了0.9962。当双亲都未知时,6个位点的累积排除率也达到了0.9674。所以利用这6个位卫星位点能够有效地确认尼罗罗非鱼各家系父本。

摘  要:
The application of microsatellite markers for parentage determination is gaining both acceptance and popularity in aquaculture. In this study,six microsatellite loci( GM024,GM435,GM459,UNH719,UNH846 and GM558) were applied in order to determine the parentage of 8 Nile tilapia families. The PCR products were genotyped using 12% polyacrylamide gel electrophoresis. The average observed heterozygosities and polymorphic information content of these six microsatellites were 0. 696 and 0. 608,respectively. The CPE( combined paternity exclusion probability) of six microsatellite loci was 0. 9962( confidence was 95%) when the mother was known. When both parents were unknown,the CPE of the six loci was 0. 9674. However,the confidence of the paternity testing was less than 80%. The results suggest that these six loci can be applied for paternity testing in Nile tilapia when the mother was known.
