不同营养类型大型真菌菌种保存培养基的筛选

摘  要:
不同营养类型的大型真菌在菌种培养和保存上对培养基质的要求有很大不同,即使同一营养类型真菌,对培养基质的要求也有较大差异.筛选适宜而通用的培养基,可为野生大型真菌种质资源保存带来便捷.本研究选取9个腐生型、8个共生型和3个兼性寄生型大型真菌菌株,采用平板培养法研究它们在改良YPD(1号)、MMN(2号)、Pachlewski(3号)、YPD-玉米粉水提液(4号)、改良PDA(5号)和YPD-平菇水提液(6号)等6种培养基上的生长情况.结果:大部分测试菌株在2号和3号培养基上长势弱,而在4号培养基上长速慢.在1号和6号培养基上,65%的菌株长速最快,90%的菌株生长浓密.因此推荐1号和6号培养基作为不同营养类型真菌菌种保存的通用培养基.

摘  要:
There are significant differences in the requirements for culture media in the cultivation and preservation of large fungi with diverse nutritional types.Even the differences in substrate requirements were also considered within the same nutritional type of mushroom.Screening for suitable and universal media can facilitate the preservation of wild macrofungal germplasm resources.In this study,9 saprotrophic,8 symbiotic,and 3 facultative parasitic strains were used for the selection of six media,including modified YPD(No.1),MMN(No.2),Pachlewski(No.3),YPD-corn steep liquor(No.4),modified PDA(No.5),and YPD-pleurotus extract(No.6).Results:65%of the strains exhibited the fastest growth rates on media No.1 and No.6,and 90%of the strains showed dense growth,while most of the strains showed the weakest growth on media No.2 and No.3,and slow growth rates on medium No.4.Therefore,media No.1 and No.6 were recommended as universal media for the preservation of strains of different nutritional types.
