烟草根际与非根际细菌的系统发育多样性研究

摘  要:
运用纯培养法和基于16S rRNA基因序列的系统发育分析对烟草根际与非根际土壤中可培养细菌多样性进行了研究。用NA、1/10NA、R2A、1/10R2A培养基分别从烟草根际与非根际土壤中分离了17株与28株细菌。16S rRNA序列分析结果表明,17株根际细菌可分为5个属6个种,7株属于厚壁菌门(Firmicutes,41.18%),4株属于变形菌门α-变形菌纲(Alphaproteobacteria,23.53%),4株属于放线菌门(Actinobacteria,23.53%),2株属于变形菌门β-变形菌纲(Betaproteobacteria,11.76%);28株非根际细菌可分为9个属17个种,15株属于厚壁菌门(Firmicutes,53.57%),6株属于放线菌门(Actinobacteria,21.43%),4株属于变形菌门β-变形菌纲(Betaproteobacteria,14.29%),2株属于变形菌门α-变形菌纲(Alphaproteobacteria,7.14%),1株属于拟杆菌门(Bacteroidetes phy.nov.,3.57%)。芽孢杆菌属(Bacillus)为优势菌属。1株细菌可能为新种或属。研究表明烟草根际与非根际土壤中含有较为丰富的微生物资源。

摘  要:
Cultureble bacteria were isolated from rhizosphere and non-rhizosphere of tabacco soil by conventional culture-dependent method and investigated by phylogenetic analysis based on 16S rRNA gene sequence to understand their diversity.Using NA,1/10NA,R2A and 1/10R2A(Difco) medium,18 and 27 bacteria strains were isolated from two sample soil respectively.The 16S rRNA result showed that the 18 rhizosphere bacteria isolates belonged to 9 species of 5 genus,of which 8 were members of Firmicutes(44.44%);4 were in Alphaproteobacteria(22.22%);4 belonged to Actinobacteria(22.22%);and 1 belonged to Betaproteobacteria(11.11%).The 27 non-rhizosphere belonged to 16 species in 9genus,and including members of Firmicutes(51.85%),Actinobacteria(22.22%),Betaproteobacteria(14.81%),Alphaproteobacteria(7.41%),and Bacteroidetes phy.nov.(3.70%).Among these bacteria,Bacillus was the dominant genus.1 bacterium isolate was candidate new species or genus.Thehere were abundant species diversity and phylogenetic diversity of bacteria in the rhizosphere and non-rhizosphere of tabacco soil.
