沙子岭猪GSG1基因克隆、生物信息学分析及组织表达

摘 要:
采用RACE技术克隆猪GSG1(Germ Cell-specific gene 1)基因cDNA1全长并进行生物信息学分析,利用实时荧光定量PCR对GSG1基因在沙子岭猪睾丸组织8个不同发育阶段(E90、D1、D30、D60、D90、D120、D150和D180)和30日龄时9个不同组织(睾丸、肌肉、小肠、肺、心脏、肾脏、肝脏、脾脏和脂肪)的表达量。结果表明:GSG1基因cDNA全长为1 257 bp,含1个长993 bp的开放阅读框,编码330个氨基酸。实时荧光定量PCR结果显示,GSG1基因在睾丸组织中的表达量最高,显著高于其余8个组织(P<0.05),且其在肌肉、小肠、肺脏、心脏、肾脏和脂肪中的表达水平极低;GSG1基因在睾丸组织的E90、D1、D30和D60这4个发育时期的表达量较低,显著低于D90、D120、D150和D180这4个时期(P<0.01)。本试验首次克隆了猪GSG1基因cDNA全长并检测了其在不同组织和不同发育阶段睾丸组织的表达,为进一步研究其调控猪睾丸发育和精子生成的机制奠定了理论基础。

摘 要:
In this study,the GSG1 cDNA full length of pig was cloned by RACE and analyzed by bioinformatics,the GSG1 gene expression levels were detected in eight different developmental stages(E90,D1,D30,D60,D90,D120,D150,D180) of Shaziling pig testicular tissue and eight different tissues(testis,muscle,lung,heart,kidney,liver,spleen,fat) of D30 period by real-time fluorescence quantitative PCR.The results showed that the full-length of porcine GSG1 cDNA was1 257 bp,with a 993 bp open reading frame encoding 330 amino acids.The quantitative PCR analysis results showed that the D30 period’s highest expression level of GSG1 mRNA was in testis,which was significantly different compared with the other tissues(P<0.05).In addition,the expression level of GSG1 mRNA in D90,D120,D150 and D180 were much higher thanthose in the other four development stages(P<0.01).The full length of GSG1 cDNA was the first cloned and the expression levels of GSG1 gene in different tissues and different developmental stages of Shaziling porcine testes were detected in this study,which provided the foundation for further understanding its functions in regulating the pig testis development and spermatogenesis.
