松树土壤根际微生物溶磷基因的克隆与表达

摘  要:
探究土壤根际微生物溶磷基因对难溶性矿物质磷的溶解作用。利用从土壤中提取宏基因组,克隆了Gab Y基因,并构建了表达载体,研究了该基因在大肠杆菌DH5α的表达对难溶矿物质磷酸Ca_3(PO_4)_2的溶解作用。结果表明,构建的Gab Y表达载体成功表达;对照组产酸能力较试验组弱,且对照组溶磷效率要低于试验组,直接从植物根际土壤微生物宏基因组中筛选溶磷基因对矿物质无机磷的溶解的研究可行。

摘  要:
To study the dissolving function of Gab Y gene on mineral inorganic phosphorus. The macro genomic DNA from soil was extracted,Gab Y gene was cloned, and then integrated into expression vector p ETDuet-1,induced the integrated vector expression in DH5α to research the dissolving function on Ca_3(PO_4)_2. The results showed Gab Y gene expression was successfully induced,and was able to promote the dissolving efficiency of inorganic phosphorus. Directly selected the mineral inorganic phosphorus soluble gene from soil macro genomic DNA is a viable method.
