核因子-κB在低氧性肺动脉高压大鼠肺组织表达的研究

摘  要:
研究核因子NF-κB在低氧性肺动脉高压大鼠肺组织表达的变化,探讨NF-κB在低氧性肺动脉高压发病机制中的作用。将20只SD大鼠随机分为正常对照组、低氧组,以常压低氧3周复制肺动脉高压模型,测量大鼠右心室Rright Ventricle(RV)和左心室+室间隔Left ventricle+septum(LV+S)重量,以RV/(LV+S)代表其右心室肥厚指数;对大鼠肺组织切片采用HE染色,经图像分析技术测定大鼠肺小动脉管壁厚度占血管外径的百分比Wall thickness/external diameter(WT%)和管壁面积占血管总面积的百分比Wall area/total vascular area(WA%)反映其肺血管重建情况。对大鼠肺组织切片采用免疫组化法观察2组大鼠肺组织NF-κB表达的变化。结果表明:1)低氧组大鼠RV/(LV+S)为(33.11±0.95)%、WT%为(22.36±2.99)%、WA%为(69.14±5.38)%,正常对照组RV/(LV+S)为(24.48±0.56)%、WT%为(13.30±2.77)%、WA%为(46.75±6.54)%,两组之间的差异有显著性(P均<0.01)。2)低氧组细支气管上皮细胞NF-κB核染色阳性细胞百分比为(29.11±1.12)%,与正常组(12.23±1.08)%比较,差异有显著性(P<0.01)。NF-κB在低氧性肺动脉高压大鼠肺组织表达增加,可能为低氧性肺动脉高压作用机制之一。

摘  要:
To investigate the expression of NF-κB in the lung tissues of chronic hypoxic rats and to explore the role of NF-κB in the pathogenesis of hypoxic pulmonary hypertension.Twenty male SD rats were randomly divided into two groups:control group,hypoxic group.The animal model of pulmonary hypertension was established by exposing the rats to normabaric hypoxic conditions for three weeks.The index of right ventricular hypertrophy RV/(LV+S) Was measured;NF-κB expression in lung tissues was observed by using immunohistochemistry.The index of wall thickness of pulmonary arteriole(WT% and WA%) was measured by a compqduterized image analyzer.1)In the hypoxic rats,RV/(LV+S) was(33.11±0.95)%,WT% was(22.36±2.99)% and WA% was(69.14±5.38)%,in the control rats,RV/(LV+S) was(24.48±0.56)%,WT% was(13.30±2.77)% and WA% was(46.75±6.54)%,there were significant differences between two groups(P<0.01).2) The percentage of positive cells for NF-κB nuclear staining in bronchiolar epithelial cells was significantly increased in the hypoxic group than that in the control group(29.11±1.12)% vs(12.23±1.08)%,P<0.01.The increase of the NF-κB expression in the lung tissues of chronic hypoxic rats.This may be one of the pathogenesis on hypoxic pulmonary hypertension.
