摘  要:
旨在探究番茄红素(lycopene, LYC)对慢性应激大鼠脾脏损伤的保护作用。选用30只体质量为180~220 g的雄性Wistar大鼠,按照试验要求随机分为3组,每组10只,分别为空白对照组(C组)、慢性应激组(CS组)、LYC干预组(CS+LYC组),其中CS组和LYC组大鼠每日9:00-15:00束缚于大鼠固定器中6 h。所有大鼠在9:00-15:00禁食禁水,余下时间各组自由采食饮水,饲养时间为21 d。第22天进行旷场试验对大鼠进行行为学检测。之后剖取脾脏,HE染色观察脾脏病理变化,TUNEL染色检测脾脏凋亡,Western blot法检测大鼠脾脏Cleaved Caspase-3、p-STAT3及STAT3蛋白表达。结果表明,与C组相比,CS组大鼠探索行为下降,出现抑郁样行为,脾脏典型组织结构不完整、白髓萎缩,边缘区部分消失,TUNEL染色表明凋亡显著增高(P<0.01),Cleaved Caspase-3和P-STAT3相对表达量显著升高(P<0.01)。LYC组大鼠与CS组相比脾脏组织未见明显病变,结构清晰完整,TUNEL染色中凋亡显著下降(P<0.01),脾脏p-STAT3的表达量明显升高(P<0.01),而Cleaved caspase-3表达量显著降低了(P<0.01)。结果提示,番茄红素可以缓解慢性应激大鼠脾脏组织损伤。

摘  要:
To investigate the protective effect of lycopene(LYC) on chronic stress-induced spleen injury in rats, thirty male Wistar rats, weighing 180-220 g, were randomly assigned to three groups: control group(C),chronic stress group(CS) and LYC group(CS+LYC).The rats of C and CS group were daily restrained stress for 6 h from 9:00 to 15:00.During the experiment(9:00-15:00),there were no food and water for all rats with or without restraint, the other time all freely eating and drinking.The feeding trial lasted for 21 d.Behavioral testing of rats via the open-field test on day 22.Then, the spleen was dissected, the pathological changes were observed by HE staining, apoptosis was detected by TUNEL,and the expression of cleaved caspase-3,p-STAT3 and STAT3 were tested by Western blot.After chronic stress, the rats in the CS group showed decreased exploratory behavior and depression-like behavior.The typical tissue structure of the spleen was destroyed, with atrophy of the white pulp and disappearance of the marginal area.The results of TUNEL showed that the rate of apoptosis increased(P<0.01).The expression of cleaved caspase-3 and p-STAT3 was increased(P<0.01).However, compared with the CS group, the spleen had a clear and intact structure with no obvious lesions in the LYC group.Meanwhile,TUNEL staining showed that the rate of apoptosis was significantly decreased in(P<0.01),the level of cleaved Caspase-3 significantly reduced,and the level of p-STAT3 was significantly increased(P<0.01) in the LYC group.These results indicated that LYC alleviates chronic stress-induced spleen damage in rats.
