正交设计优化棉花黄萎病菌ISSR-PCR反应体系

摘  要:
采用正交试验设计方法,对ISSR的影响因素进行研究,从Taq酶、Mg2+、dNTP和引物4因素3水平进行优化试验,筛选并建立了适合棉花黄萎病菌的ISSR-PCR最佳反应体系,即25μL的反应体系中含有1×PCR buff-er,200μmol/L dNTP,0.75μmol/L引物,150 mmol/L Mg2+,1U TaqDNA聚合酶和40 ng模板DNA。

摘  要:
In this study,the orthogonal design was used to optimize ISSR amplification system on Verticillium dahliae of cotton in four factors(Taq poly-merase,Mg2+,dNTP,primer) at three levels respectively.A suitable ISSR-PCR reaction system was established,namely 25 μL reaction system containing 1×PCR buffer,200 μmol/L dNTP,0.75 μmol/L primer,150 mmol/L Mg2+,1UTaq DNA poly-merase and 40ng DNA templet.
