海门山羊、徐淮山羊两个产羔性状候选基因的遗传多样性及其与繁殖力的关联分析

摘  要:
为海门山羊和徐淮山羊繁殖力标记辅助选择提供依据,以海门山羊(113只)和徐淮山羊(78只)为试验动物,将FSH-β和GDF-9基因第2外显子作为影响山羊产羔性状的候选基因,对其第2外显子进行扩增、测序、序列比对以及PCR-SSCP检测,探讨FSH-β和GDF-9基因第2外显子的遗传特征,寻找与产羔数相关的遗传标记。结果表明:FSH-β基因在海门山羊中有AA、AB、AC和CC 4种基因型,基因型频率分别为0.477 9、0.017 7、0.407 1和0.097 3。FSH-β基因在徐淮山羊中有AA、AB和AC3种基因型,基因型频率分别为0.397 7、0.477 3和0.125 0。GDF-9基因在海门山羊和徐淮山羊中有GG和GH 2种基因型,在海门山羊中基因型频率分别为0.539 8和0.460 2,在徐淮山羊中基因型频率分别为0.784 1和0.215 9。FSH-β基因在海门山羊产羔数中AC基因型最小二乘均值比AA型多0.160只,比AB型多0.250只,比CC型多0.300只,各基因型产羔数无显著差异。GDF-9基因在海门山羊产羔数中GH基因型最小二乘均值比GG型多0.179只,各基因型产羔数无显著差异。试验初步认为FSH-β和GDF-9基因不是影响海门山羊多胎性能的主效基因。

摘  要:
FSH-βand GDF-9genes as fecundity candidate genes were studied in Haimen goats(113ear sample)and Xuhuai goats(88ear sample)by PCR-SSCP technology.The results of the study would provide the theory basis for marker-assisted selection in high prolificacy of Haimen goats and Xuhuai goats.According to the performance,there were4 different genotypes of FSH-βgene in Haimen goats,in which the AA,AB,ACand CC genotype frequencies were0.477 9,0.017 7,0.407 1and 0.097 3respectively,and the AA,ABand ACgenotype frequency of Xuhuai goats were0.397 7,0.477 3and 0.125 0respectively.But for GDF-9genes,there were two different genotypes in both Haimen goats and Xuhuai goats.The genotype frequencies of GGand GH were 0.539 8,0.460 2and 0.784 1,0.215 9respectively in Haimen goats and Xuhuai goats.The Haimen goats ewe with genotype AChad 0.160 3,0.250 0and 0.300 0lambs more than those with genotype AA,ABand CCrespectively,which differed not significantly.The Haimen goats ewe with genotype GH had 0.178 8(P>0.05)lambs more than those with genotype GG,which differed not significantly.The study showed that FSH-βand GDF-9genes were not major genes that can effect high fertility of goat.
