华北大黑鳃金龟气味受体OrCo基因的克隆及序列分析

摘  要:
嗅觉受体OrCo(olfactory receptor coreceptor)在不同昆虫体内高度保守,克隆华北大黑鳃金龟[Holotrichiaoblita(Falderman)]OrCo基因可以为进一步探索受体在气味识别过程中所起的作用和功能提供基础。本研究利用RT-PCR和RACE方法,克隆获得华北大黑鳃金龟OrCo受体基因cDNA全长序列。将该基因命名为Hobl/Or-Co。序列分析结果显示,Hobl/OrCo全长共1 598bp,开放阅读框全长1 434bp,编码477个氨基酸,序列中有7个跨膜区和高度保守的C端区域。Hobl/OrCo基因的氨基酸序列与近缘种铅灰齿爪鳃金龟嗅觉受体的同源性高达95.39%,与已经报道的其他昆虫的嗅觉受体同源性在76%以上,特别是在C端几乎完全一致。

摘  要:
cDNA cloning of an unusually highly conserved olfactory receptor OrCo(olfactory receptor coreceptor) of Holotrichia oblita(Falderman) was conducted in order to provide the basis for exploring the functions of the olfactory receptor in odorant recognition.The OrCo receptor gene was cloned from the antennae of H.oblita by using reverse transcription polymerase chain reaction(RT-PCR) and rapid amplification of cDNA ends(RACE).The full-length cDNA sequence of OrCo from H.oblita was named as Hobl/OrCo.Sequence analysis revealed that the transcript consisted of 1 598 nucleotides and the open reading frame(ORF) encoded a peptide of 477 amino acids with 7 putative transmembrane domains and highly conserved carboxy-terminal.Alignment analysis indicated that the deduced amino acid sequence of OrCo gene from H.oblita shared high homologies with similar olfactory receptors from other insects(>76%),especially in the C-terminal,the homology with the receptor from the close sibling species Holotrichia plumbea reached 95.39%.
