鸽肉营养成分与脂肪沉积相关基因的研究

摘  要:
为了明确卡努鸽肉中营养成分及与脂肪沉积相关基因的表达,试验选择同批孵化、体况较好的3,12月龄雌、雄鸽各10只,屠宰后取胸大肌检测主要营养成分(水、粗蛋白、粗脂肪)及雄鸽肉中脂肪酸含量,并用RT-qPCR技术分析了与鸽肉脂肪沉积相关基因LPL、PLIN1、FASN和PPARγ的表达规律。结果表明:雌、雄鸽3月龄肉中水分、粗蛋白含量均显著高于12月龄(P<0.05),粗脂肪含量则显著低于12月龄(P<0.05);12月龄雌、雄鸽肉中水、脂肪和蛋白含量均差异不显著(P>0.05);3月龄鸽肉中肉豆蔻酸、棕榈酸、硬脂酸等主要饱和脂肪酸含量显著高于12月龄(P<0.05),而油酸、亚油酸等不饱和脂肪酸含量显著低于12月龄(P<0.05)。鸽肉中LPL基因mRNA表达量显著高于其他三种基因(P<0.05)。LPL、PLIN1和FASN基因表达量与肌内脂肪含量呈显著或极显著正相关(P<0.05或P<0.01),而PPARγ基因表达量与肌内脂肪含量呈显著负相关(P<0.05)。说明LPL基因的表达与鸽肉脂肪沉积关系更为密切,其中12月龄卡努鸽肉中脂肪酸含量更符合屠宰标准。

摘  要:
In order to detect the relationship between the nutrient content of carneau meat and the expression of intramuscular fat accumulation related genes, the main nutrition components(water, protein and fatty) in the carneau muscle of different gender at 3 and 12 months and the content of fatty acids in male pigeon meat were detected in this study. Real-time fluorescence quantitative PCR was used to detect the mRNA levels of genes associated with pigeon fat deposition, including Lipoprotein lipase(LPL), Perilipin(PLIN1), Fatty acid synthase gene(FASN), and Peroxi-some proliferator-activated receptors γ(PPARγ). The results showed that the contents of water and protein in male and female pigeons of 3 months old were significantly higher than that of 12 months old(P<0.05), and the content of crude aunt was opposite(P<0.05); there was no significant difference in water, fat and protein content between males and females carneau meat aged 12 months(P>0.05); the content of the major saturated fatty acids(myristic acid, palmitic acid and stearic acid) of 3 months old pigeon meat was significantly higher than that in 12-month-old pigeon meat(P<0.05), while those of unsaturated fatty acids(oleic acid and linoleic acid) were opposite(P<0.05). The mRNA expression of LPL gene in pigeon meat was significantly higher than the other three genes(P<0.05). The mRNA expression of LPL, FASN and PLIN1 was significantly positively correlated with intramuscular fat content(P<0.05 or P<0.01), while PPARγ was significantly negatively correlated with intramuscular fat content(P<0.05). It indicated that the expression of LPL gene was more closely related to the fatty deposition of carneau meat, and the fatty acid content of 12-month-old carneau meat was more in line with the slaughter standard.
