杏鲍菇菇头多糖的结构鉴定及生物活性评价

摘  要:
采用热水浸提法从杏鲍菇菇头中提取粗多糖,采用凝胶渗透色谱-示差折光指数-十八角激光散射联用技术测定粗多糖的分子质量分布,气相色谱法测定单糖组成,气相色谱-质谱联用法测定糖苷键链接方式,2,2’-联氨-双-3-乙基苯并噻唑啉-6-磺酸(2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid),ABTS)法测定抗氧化性,3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐法测定体外抗肿瘤活性,动物实验法检测体内抗肿瘤活性。结果表明:该粗多糖主要由葡萄糖组成(占84.4%);其分子质量分布较广,4个主峰分子质量分别为3.816×10~7、2.010×10~6、4.572×10~5 Da和1.849×10~5 Da;糖苷键主要链接方式为1,3-糖苷键(占42.7%)和1,6-糖苷键(占35.5%);当质量浓度为5 mg/mL时,粗多糖对ABTS阳离子自由基的抑制率为70.9%。动物实验表明,以200 mg/(kg·d)剂量连续灌胃12 d后,粗多糖对荷瘤小鼠(宫颈癌U14)的抑瘤率可达39.84%。

摘  要:
Crude polysaccharide(CP) was obtained by hot water extraction from the stalk residue of Pleurotus eryngii. The molecular mass distribution of CP was assayed by the combined use of gel permeation chromatography(GPC), differential refraction detector(RID), and multi-angle light scattering(MALLS). The monosaccharide composition was assayed by gas chromatography(GC). The glycosyl linkages were analyzed by gas chromatography-mass spectrometry. The antioxidant activity was measured by the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)(ABTS) radical cation assay. The antitumor activity was assayed by both in vitro 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) and in vivo methods. Results showed that glucose was the predominant monosaccharide(84.4%) of CP; the molecular mass of CP was widely distributed and mainly composed of 4 peaks(i.e. 3.816 ×10~7, 2.010 ×10~6, 4.572 ×10~5 and 1.849 ×10~5 Da). The glycosyl linkages mainly consisted of 1,3-linked(42.7%) and 1,6-linked(35.3%) glucose residues. At a concentration of 5 mg/mL, the crude polysaccharides exerted 70.9% scavenging rate against ABTS radical cation. In addition, the in vivo test showed that CP could significantly inhibit 39.84% of the growth of transplantable U14 cervical cancer in mice at a dose of 200 mg/(kg·d) after 12 days of intragastric administration.
