米曲霉碱性蛋白酶基因的前导区对其在毕赤酵母中表达的必要性

摘 要:
为探讨前导区对米曲霉碱性蛋白酶基因在毕赤酵母中表达的必要性,本试验首先将米曲霉碱性蛋白酶的成熟肽编码基因(alp-m)和表达载体pPIC9K双酶切后连接,得到重组表达载体pPIC9K/alp-m。然后将其线性化后电转化毕赤酵母GS115,筛选阳性转化子进行甲醇诱导表达,并进行SDS-PAGE分析和碱性蛋白酶活力测定,同时在转录水平上对目的基因进行了Northern blot检测。将得到的结果与以前研究中得到的带有前导区的米曲霉碱性蛋白酶成熟肽基因在毕赤酵母中的表达结果进行对比。结果表明:如果没有前导区,米曲霉碱性蛋白酶成熟肽基因在毕赤酵母中表达,无论在胞内和胞外均检测不到目的蛋白。由此可见,米曲霉碱性蛋白酶基因在毕赤酵母中表达时必须有前导区的参与。

摘 要:
To study the essentiality of its pro-peptide sequence when Aspergillus oryzae alkaline protease (alp) gene was expressed in Pichia pastoris, the cDNA encoding mature protein of alkaline protease from A. oryzae without pro-peptide sequence was cloned into the vector pPIC9K. Then the recombinant plasmids pPIC9K/alp-m were linearized and transformed into P. pastoris GS115 competent cells. After the positive transformants were induced by methanol, the analysis of SDS-PAGE and alkaline protease activity were carried out. Besides, Northern blot was used to detect the expression of alp-m gene in transcription level. The results obtained with above methods were compared with the result obtained in our previous study where mature protein sequence of alkaline protease from A. oryzae with pro-peptide was expressed in P. pastoris. Without pro-peptide sequence, no alkaline protease was detected either in the cell or out the cell when alp gene was expressed in P. pastoris. Thus, pro-peptide sequence is required when alp gene is expressed in P. pastoris.
