奶牛β-酪蛋白基因的克隆及序列分析

摘  要:
利用高保真PCR技术扩增了黑白花奶牛β-酪蛋白基因(CSN2)5’和3’调控成分,纯化PCR产物与pMD 19-TVector亚克隆后酶切鉴定和测序。结果表明:克隆片断与奶牛β-酪蛋白基因相应区域同源性分别为97.0%和99.0%,并且包含有大部分的CSN2表达核心调控序列和多个转录、翻译因子的结合位点,获得了黑白花奶牛β-酪蛋白基因调控区的克隆。为构建乳腺特异表达载体,指导外源基因在转基因动物乳腺内高效表达奠定了基础。

摘  要:
The high-fidelity PCR had been used to amplify 1.8 kb and 1.1 kb 5' and 3' regulatory sequences of bovine casein gene in mammary grand,after the of the PCR production,it was cloned into pMD 19-T vector,and then restriction enzyme was used for digestion and sequence analysis.Results of software indicated that the homology of fragments with the correspondingly region of bovine beta-casein was 97.0% and 99.0% respectively,most of the vital regulatory elements and many factors binding sites were found in the two fragments.It was also described that the restructuring carrier had succeeded in cloning the area of control casein gene of 5' and 3' It could be concluded that the constructing expression system of bovine mammary gland-specific expression vector and lay the foundation on instructing extraneous source gene highly effective expression in transferred animal mammary gland.
