NaN3诱变对赤小豆种子萌发与幼苗抗氧化系统的影响

摘  要:
叠氮化钠(NaN3)诱变是获得基因水平变异体的重要途径,了解叠氮化钠处理时间和处理浓度等基本条件是进行诱变育种的前提.本研究采用0.0,0.2,0.6,1.0,1.4,1.8mmol/L的叠氮化钠溶液浓度对赤小豆种子进行诱变处理,处理时间为16,20,24h.通过发芽率及相对发芽率确定叠氮化钠诱变赤小豆的适宜浓度和时间(半致死剂量LD50).此外,还对赤小豆M1代幼苗中超氧化物歧化酶(SOD)、过氧化物酶(POD)以及过氧化氢酶(CAT)活性进行检测,探究叠氮化钠对赤小豆幼苗抗氧化系统的影响.结果表明:叠氮化钠对赤小豆诱变处理时间为20h时的半致死浓度为1.0mmol/L.叠氮化钠对赤小豆幼苗的SOD、POD活性表现为低浓度促进,高浓度抑制;对赤小豆幼苗的CAT活性有促进作用.当叠氮化钠浓度为1.0mmol/L时,赤小豆幼苗的SOD活性最高;而浓度为0.6mmol/L时,POD活性最高.

摘  要:
Sodium azide(NaN_3)mutagenesis is an important way to obtain gene-level variants,understanding the basic conditions such as the treatment time and concentration of sodium azide is the precondition for mutagenesis breeding.In this study,we treated rice bean seeds by sodium azide solution on the concentrations of 0,0.2,0.6,1.0,1.4 and 1.8 mmol/L(solvent for phosphate buffer solution,pH=3)with treatment time of 16 h,20 hand 24 hrespectively.Though testing and analyzing germination rate and relative germination rate,we tested the optimum mutagenic dose concentration(half lethal dose LD 50)and suitable time of mutation.In addition,superoxide dismutase(SOD),sodium peroxidase(POD)and catalase(CAT)activity of the seedlings of rice bean M1 generation were detected,explore sodium azide effects on antioxidant system of rice bean seedlings.The results showed that the semi-lethal concentration of sodium azide on rice bean was 1.0 mmol/L and the time was 20 h.The activities of SOD and POD in the seedlings of rice bean were promoted by low concentration and inhibited by high concentration.While the CAT activity of red bean seedlings was always promoted.When the concentration of sodium azide was 1.0 mmol/L,the SOD activity of rice bean seedlings was the highest,while the concentration was 0.6 mmol/L,POD activity was the highest.
