摘  要:
[目的]建立云南小粒咖啡中绿原酸、葫芦巴碱、咖啡酸、D-(-)-奎宁酸含量的测定方法。[方法]采用Venusil ASB C18为色谱柱,甲醇-0.1%磷酸水为流动相,等度洗脱,流速0.9 m L/min,柱温35℃,检测波长:绿原酸和咖啡酸330 nm,葫芦巴碱和D-(-)-奎宁酸210 nm。[结果]绿原酸、葫芦巴碱、D-(-)-奎宁酸、咖啡酸分别在14.6~146.0μg/m L(r=1.000 0)、10.2~102.0μg/m L(r=1.000 0)、11.6~116.0μg/m L(r=0.999 8)、0.499 5~4.995 0μg/m L(r=0.999 8)的浓度范围内与峰面积线性关系良好,加标回收率为93.28%~97.46%。[结论]该方法简便、准确、重复性好,可用于云南小粒咖啡中绿原酸、葫芦巴碱、咖啡酸和D-(-)-奎宁酸含量的测定。

摘  要:
[Objective]To establish a method for the determination of chlorogenic acid,trigonelline,caffeic acid and D-(-)-quinic acid in Arabica of Yunnan by HPLC.[Methods]The HPLC conditions were ascertained as follows: Venusil ASB C18 chmatgraphic column,mobile phase methanol plus 0.1% phosphoric acid,isometric elution; flow rate 0.9 m L/min; column temperature 35 ℃; determination wavelength: chlorogenic acid and caffeic acid with 330 nm,trigonelline and D-(-)-quinic acid with 210 nm.[Results]There were linear relationships between the peak area and contents of determination of chlorogenic acid,trigonelline,caffeic acid and D-(-)-quinic acid in the ranges of 4.6 ~ 146.0 μg/m L(r =1.000 0),10.2 ~ 102.0 μg/m L(r = 1.000 0),11.6 ~ 116.0 μg/m L(r = 0.999 8),0.499 5 ~ 4.995 0μg/m L(r = 0.999),respectively.The extraction recoveries varied between 93.28%-97.46%.[Conclusion]The method was simple,accurate,reliable,and can be used to detect chlorogenic acid,trigonelline,caffeic acid and D-(-)-quinic acid in coffea arabica.
