金黄色葡萄球菌小菌落突变株体外感染奶牛乳腺上皮细胞的研究

摘  要:
为了研究金黄色葡萄球菌小菌落突变株对正常奶牛乳腺上皮细胞的侵袭作用,试验采用普通光学和扫描电子显微镜观察的方法对临床分离的一株金黄色葡萄球菌小菌落突变株及金黄色葡萄球菌质控菌株ATCC25923体外感染培养的奶牛乳腺上皮细胞引起的细胞形态变化及细菌侵入细胞的过程进行了研究。结果表明:金黄色葡萄球菌分离菌株与质控菌株ATCC25923在体外感染乳腺上皮细胞3 h后出现凋亡现象,表现为细胞体积缩小,核质浓缩,核膜核仁破碎,胞膜有小泡状形成。说明金黄色葡萄球菌小菌落突变株对奶牛乳腺上皮细胞具有致病性,但致病力远低于金黄色葡萄球菌质控菌株ATCC25923,金黄色葡萄球菌小菌落突变株可以在奶牛乳腺上皮细胞中持续存在,在细胞中有一定存活,与金黄色葡萄菌株质控菌株ATCC25923相比对细胞的侵袭能力较低。

摘  要:
In order to study the invasion effect of Staphylococcus aureus small mutant on normal bovine mammary epithelial cells,using common light microscopy and scanning electron microscopy,a clinical isolated mutant and the quality control strain ATCC25923 of Staphylococcus aureus were tested in this experiment to infect bovine mammary epithelial cells for the ovservation of morphologic changes on epithelial cells and of invasion process of bacteria to cells. The results showed that both the mutant and ATCC25923 quality control strains led to apoptosis in mammary epithelial cells 3 h post infection,with cell volume decreased,cell nucleolus concentrated,nuclear membrane and nucleolus broken,and also with small vesicle formation on cell membrane. The results suggested that the Staphylococcus aureus small mutant was pathogenic to bovine mammary epithelial cells but far below the pathogenicity of the quality control strain ATCC25923. The small mutant could exist persistently in bovine mammary epithelial cells,with a certain proportion of bacteria survival in the cells. This small mutant had a lower invasion ability to the cells compared with the quality control strain ATCC25923.
