HPLC-ELSD法测定木薯块根中蔗糖、葡萄糖、果糖的含量

摘  要:
可溶性糖为木薯块根淀粉的合成提供底物和能量,准确测定其含量有助于进一步了解木薯块根淀粉积累过程。本研究以水提法提取木薯块根中的可溶性糖,采用高效液相色谱-蒸发光散射检测法(HPLCELSD)测定样品中果糖、葡萄糖、蔗糖的含量。以Waters-NH2色谱柱结合Alltech 3300 ELSD蒸发光检测器进行糖含量的测定,流动相为乙腈/水(70/30,V/V),含0.1%的氨水。果糖、葡萄糖和蔗糖的分离效果较好,在1~25μg范围内线性关系良好,精密度RSD分别为0.31%、0.22%和1.42%,回收率分别为97.62%、98.03%、96.30%,样品稳定性实验的RSD分别为0.88%、0.48%和1.05%。实验结果表明HPLC-ELSD法是分析木薯块根中的蔗糖、葡萄糖、果糖含量的有效技术,可为研究淀粉含量不同的木薯品种在糖代谢过程中的差异奠定基础。

摘  要:
The soluble sugar provides the substrate and energy for the synthesis of starch in cassava tuber root,and the accurate determination of its content may helps to further understand the starch accumulation process. In this research, soluble sugars in cassava tuber were extracted by water. The content of fructose, glucose, sucrose in sample was determined using high-performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD). Sugars were separated on a Waters-NH_2 column with a 70:30(V/V) mixed solution of acetonitrile and 0.1% ammonia water as moble phase and detected by Alltech 3300 ELSD evaporative light detector. Fructose,glucose and sucrose could be completely separated. The linear relations of fructose, glucose and sucrose were good in the range of 1~25 μg, and the precision(RSD %) was 0.31%, 0.22% and 1.42%, respectively; the recoveries were 97.62%, 98.03% and 96.30%, respectively; the precision(RSD %) of sample stability was 0.88%, 0.48% and 1.05%, respectively. The results showed that HPLC-ELSD method is an effective technique for the analysis of sucrose, glucose and fructose content in cassava tuber roots, which could lay a foundation for the study of differences in sugar metabolism of cassava cultivars with different starch contents.
