创伤弧菌浸泡感染对孔雀鱼的影响

摘  要:
为建立创伤弧菌(Vibrio vulnificus)的感染模型,了解创伤弧菌对孔雀鱼(Poecilia reticulate)的致病力以及创伤弧菌感染对孔雀鱼免疫相关因子表达的影响。将孔雀鱼分为4组,分别以创伤弧菌FZ03-5X浓度为3.3×108、3.3×107、3.3×106和0 CFU/mL的菌液浸泡感染5 h,检测感染15 d后3.3×107CFU/mL浸泡组和生理盐水对照组鳃免疫相关因子和凋亡相关因子的表达水平。结果表明,4个浓度组孔雀鱼的死亡率分别为40%、18.33%、13.33%和6.67%。感染15 d后,肿瘤坏死因子(TNF)、Mx因子、与细胞凋亡相关的凋亡因子FAS和含半胱氨酸的天冬氨酸蛋白水解酶(caspase)的表达水平显著高于生理盐水对照组(P<0.05)。试验结果说明,创伤弧菌可通过浸泡的方式使孔雀鱼致病,孔雀鱼因创伤弧菌感染产生的炎症反应可持续15 d。

摘  要:
The purpose of this study is to establish an infection model of Vibrio vulnificus, to observe the pathogenicity of V. vulnificus to guppies and to understand the effect of V. vulnificus infection on the expression of immune related genes of guppies. Guppies were divided into 4 groups, and immersed, respectively, in the V. vulnificus FZ03-5 X liquid with a concentration of 3.3×108, 3.3×107,3.3×106 and 0 CFU/mL for 5 h. The expression levels of immune-related factors and apoptosis-related factors of the fish immersed in 0 and 3.3×107 CFU/mL V. vulnificus liquid were determined at 15 days after infection. The results showed that the mortality of 3.3×108,3.3×107, 3.3×106 and 0 CFU/mL immersed group was, respectively, 40%, 18.33%,13.33% and 6.67% after 15-day infection. And the expression level of the tumor necrosis factor(TNF), Mx factor, apoptosis-related apoptosis factor FAS and cysteine-containing aspartate proteolytic enzyme(caspase) of the fish immersed in 3.3×107 CFU/mL V. vulnificus liquid was significantly higher than that of the control fish(P<0.05). The results reveal that V. vulnificus can cause disease of guppies by immersion, and the inflammatory reaction of guppies to V. vulnificus infection can last for 15 days.
