激素对菘蓝叶片组织培养的影响

摘  要:
[目的]为提高菘蓝的生产效率、降低其生产成本提供技术支持。[方法]以菘蓝无菌苗的叶片为外植体,利用正交设计研究6-BA和NAA对菘蓝叶片分化的影响。[结果]处理6-BA 1.5 mg/L+NAA 0.6 mg/L和6-BA 2.0 mg/L+NAA 0.2 mg/L的不定芽分化率均为93.75%,分化系数分别为40.0和38.75。NAA对叶片分化率的影响最大,其与6-BA的交互作用较强,6-BA的影响相对较小。菘蓝叶片分化的最佳培养基为MS+6-BA 2.0 mg/L+NAA 0.6 mg/L。IBA和NAA对菘蓝试管苗生根均有一定的促进作用。单独使用0.1 mg/LNAA的生根率达97%以上且根的质量较好,而过高浓度抑制生根。IBA对菘蓝生根的促进作用不如NAA,生根率较低且根的质量较差。菘蓝生根的适宜培养基为MS+NAA 0.1 mg/L。[结论]该研究为菘蓝的组织培养和快速繁殖奠定了初步基础。

摘  要:
[Objective] The research aimed to provide technical support for enhancing the production efficiency of Isatis indigotica and reducing its production cost.[Method] With leaves of aseptic I.indigotica seedlings as explants,the effects of 6-BA and NAA on the differentiation of I.indigotica leaves were researched by orthogonal design.[Result] The differentiation rates of adventitious buds in both the treatments 6-BA 1.5 mg/L+NAA 0.6 mg/L and 6-BA 2.0 mg/L+NAA 0.2 mg/L were 93.75% and their differentiation coefficients were 40.0 and 38.75 resp.The effect of NAA on the differentiation rate of leaves was greatest,its interaction with 6-BA was stronger and the effect of 6-BA was relatively less.The optimum medium for the differentiation of I.indigotica leaves was MS+6-BA 2.0 mg/L+NAA 0.6 mg/L.Both IBA and NAA had some promoting effect on the rooting of I.indigotica plantlets.When 0.1 mg/L NAA was used individually,the rooting rate was higher than 97% and the quality of roots was better,but the higer concn.would inhibit rooting.The promotion of IBA on the rooting of I.indigotica was inferior to that of NAA,its rooting rate was lower and the quality of roots was worse.The suitable medium for the rooting of I.indigotica was MS+NAA 0.1 mg/L.[Conclusion] The research layed a primary foundation for the tissue culture and rapid propagation of I.indigotica.
