摘 要:
主要组织相容性复合体(major histocompatibility complex,MHC)-四聚体(tetramer)技术是模拟生物体内抗原提呈原理,将4分子的生物素化的MHC-抗原肽单聚体与1分子荧光标记的链霉亲合素连接成四聚体复合物,定向、定量对抗原特异性T细胞进行分析及检测。该技术已被用于疾病诊断、疾病防控和相关科学研究,尤其在病毒、癌症、自身免疫疾病方面具有广阔的应用前景,是当今免疫学和分子生物学领域研究的热点之一。文章就MHC的分子结构基础,四聚体的制备、优化及其应用做一综述。

摘 要:
MHC tetramer technology was based on antigen-presenting principle in vivo. Tetramer complexs were made up of four molecules of biotinylated MHC-antigen peptide monomeric molecules with one molecule of streptavidin,which could be used to detect and analyze antigenspecific T cells directionally and quantitatively. The technology had been applied in disease diagnosis,disease prevention and control,and related scientific research,particularly,in terms of viruses,cancer,autoimmune diseases. It was one of the hotspots in current immunology and molecular biology research. In this paper,the molecular structure of MHC,preparation,optimization,and application of the tetramer were reviewed.
