兽用口服地克珠利纳米乳递药系统的构建及其性质考察

摘 要:
为了构建稳定的兽用口服地克珠利纳米乳递药系统,通过检查地克珠利在不同油相中的溶解性,以及最佳油相在不同表面活性剂中的溶解性,筛选出最佳油相和最佳表面活性剂,利用伪三元相图优选出最佳助表面活性剂、表面活性剂-助表面活性剂最佳体积比(Smix)和Smix-油相最佳体积比,并结合冷-热循环试验、离心试验和冻-融试验考察稳定性,以确定地克珠利纳米乳递药系统的最佳处方;鉴别最佳处方样品类型,同时测定不同稀释比例条件下地克珠利纳米乳的粒径分布、Zeta电位,并利用透射电镜观察地克珠利纳米乳递药系统的微观形态.结果显示,构建地克珠利纳米乳递药系统的最佳油相为大豆油,最佳表面活性剂为EL-40,最佳助表面活性剂为PEG400,Smix为1∶1,Smix-油相最佳体积比为7∶3(1∶0.43);构建的地克珠利纳米乳递药系统为水包油(O/W)型,稳定性良好,经10、100、1 000倍蒸馏水稀释后的平均粒径分别为15.81、15.85、22.44 nm,其Zeta电位分别为-3.180、-1.620、-0.786 mV,在电镜下观察其外观呈球形且大小均匀无黏连.综上,构建的地克珠利纳米乳递药系统稳定性良好,可与水以任意比例混溶.

摘 要:
In order to construct a stable animal oral diclazuril nanoemulsion delivery system,the best oil phase and surfactant were screened out by checking the solubility of diclazuril in different oil phases and the best oil phase in different surfactants.The best cosurfactant,the best volume ratio of surfactant-cosurfactant(Smix) and the best volume ratio of Smix-oil were optimized by pseudoternary phase diagram,and combining cold-heat cycle test,centrifugal test and freeze-thaw test,the stability was investigated to determine the best prescription.The best prescription sample type was identified and the particle size distribution and Zeta potential were determined at different dilution ratios.The morphology of nanoemulsion was observed by transmission electron microscopy.The results showed that the optimum oil phase of diclozuril nanoemulsion was soybean oil,the optimum surfactant was EL-40,the optimum cosurfactant was PEG400,Smix was 1∶1,and the best volume ratio of Smix-oil was 7∶3(1∶0.43).The prepared diclozuril nanoemulsion was oil in water(O/W) type and had good stability.After dilutioned by 10,100 and 1 000 times distilled water,the average particle size were 15.81,15.85,22.44 nm,and Zeta potential were-3.180,-1.620,-0.786 mV,respectively.The appearance of diclozuril nanoemulsion was spherical with uniform size and no adhesion.The results showed that the nanoemulsion prepared in the study with good stability and ability to mixed with water in any proportion.
