人工种植与道地野生的金莲花药用品质比较及超声波提取多糖工艺优化

摘  要:
为测定人工种植的金莲花(Trollius chinensis Bunge)中多糖提取率和探索超声波辐射辅助下多糖提取的最佳工艺,并通过人工种植与道地野生的金莲花中多糖提取率的比较,实施对人工种植金莲花的药用品质评价;试验在超声波辐射下,用水提醇沉法提取金莲花多糖、三氯乙酸法除去蛋白、苯酚-硫酸法测多糖含量,通过单因素试验和正交试验,比较了超声波辐射条件下超声波辐射功率、超声波辐射时间、料液比、提取温度对多糖提取率的影响。结果人工种植的金莲花多糖提取率为2.94%,最佳提取工艺条件是超声波辐射功率300 W、料液比1∶25、超声波辐射时间30 min、提取温度80℃。这个结果显示人工种植的金莲花多糖含量较高,完全能满足药用需求。

摘  要:
Polysaccharide content in artificial planting globeflower(Trollius chinensis Bunge) was determined and ultrasonic extraction technology was optimized. Through comparing polysaccharide of artificial planting and authentic wild globeflower the medicinal quality of artificial planting globeflower was evaluated. Under ultrasonic radiation,polysaccharide of globeflower was extracted by water and deposited by ethanol. Deproteinized by TAC the content of polysaccharides was determined with phenol-sulfuric acid. Effects of ultrasonic irradiation and ultrasonic power, ultrasonic radiation time, temperature, liquid ratio of polysaccharide extraction rate were studied with single factor test and orthogonal test. Polysaccharide content of artificial planting globeflower was 2.94%. The optimal conditions were Ultrasonic power of 300 W, the liquid material of 1∶25,ultrasonic irradiation time of 30 min, temperature of 80 ℃. Polysaccharide content of artificial planting globeflower was high and fully met the medicinal requirement.
