采用批实验方法研究了一株耐冷约氏不动杆菌DBP-3在低温和低碳氮比条件下对氨氮的去除特性。结果表明,将菌株DBP-3接种于乙酸钠和氨氮分别为唯一碳源和氮源的基础培养基中,10℃和碳氮比为6时培养60 h后,培养液的OD600可达0.658,氨氮的去除率达到89.7%,总有机碳的浓度下降95%,硝态氮和亚硝态氮的累积浓度分别为12.5、6.8 mg·L-1,并且在整个培养过程中都出现了先增加后下降的趋势。菌株DBP-3能大量消耗乙酸盐进行生长和氧化氨氮,其次为琥珀酸和柠檬酸,对葡萄糖的利用程度非常低,并且几乎不能利用蔗糖。碳氮比低于8时,随碳氮比的增加,氨氮的去除率逐渐增加,碳氮比为4和6时,增加氨氮的初始浓度可明显提高其去除率,在氨氮浓度为200 mg·L-1时,碳氮比4和6的培养体系中氨氮的去除率可分别达到58.12%和83.38%。溶解氧水平对菌株DBP-3的氨氮去除能力具有一定的影响,当溶解氧水平为3.1 mg·L-1和4.2 mg·L-1时,菌株对氨氮的去除率仍然可分别达到28.42%和50.36%。在10℃和碳氮比为6时,氨氮的浓度从5增加至80 mg·L-1,培养体系中氨氮的浓度均可降低88%以上。以上研究结果可为菌株DBP-3应用于低温和低碳氮比富营养化水体的生物脱氮处理研究提供理论依据。

Under the conditions of low temperature and C/N ratio,the removal of NH+ 4-N by a psychrotolerant Acinetobacter johnsonii DBP3 through the batch testing experiment.Results showed that OD600 values of cultural media was 0.658 and the removal rate of NH+ 4-N and the concentrations of nitrate and nitrite were 89.7%,12.5,6.8 mg·L-1,respectively after 60 h of cultivation under conditions of 10 ℃ and C/ N ratio 6 when sodium acetate and NH+ 4-N were used as the sole carbon and nitrogen sources,respectively.In addition,during the cultivation,the concentration of nitrate and nitrite slowly increased followed a decreasing trend.Strain DBP-3 used acetate as carbon and energy source substantially to grow and remove NH+ 4-N,followed by citrate and succinate,but the using degree of glucose by strain DBP-3 was very low and it could not utilize sucrose.The removal rate of NH+ 4-N increased with the increasing of C/N ratio when the C/N ratio was lower than 8.The increased concentration of NH+ 4-N significantly enhanced the removal rate of NH+ 4-N with C/N ratios of 4 and 6.The removal rates of NH+ 4-N were 58.12% and 83.38% in cultural media with C/N ratios of 4 and 6,respectively,when the initial concentration of NH+ 4 N was 200 mg·L-1.The dissolved oxygen level in the cultural media affected the removal of NH+ 4-N,but the removal rate of NH+ 4-N by strain DBP-3 could still be 28.42% and 50.36% with the shaking speed of 0 and 40 r·min-1,respectively.The average NH+ 4-N removal rate in the cultural media by strain DBP-3 was higher than 88% when the initial concentration of ammonium ranged from 5 to 80 mg·L-1under conditions of 10 ℃ and C/N ratio 6.These data may be helpful to support the use of strain DBP-3 in the nitrogen removal of eutrophic water bodies with low temperatures and C/N ratio.
