摘 要:
云南省是我国大蚕蛾科(Saturniidae)绢丝昆虫资源最为丰富的地区之一。利用DNA条形码技术对采自云南省楚雄地区的一种野生绢丝昆虫进行分子鉴定,并调查其茧质性状。提取该野生绢丝昆虫的蛹基因组DNA,以昆虫线粒体细胞色素C氧化酶亚基Ⅰ基因(mt DNA COⅠ)片段通用引物PCR扩增,获得一段长度为574 bp的mt DNA COⅠ基因片段(Gen Bank登录号:KR812471)。基于mt DNA COⅠ基因序列比对,该野生绢丝昆虫与柞蚕属(Antheraea)的明目大蚕蛾(A.frithi)同源序列的相似度为99.47%。应用Kimura-2-Parameter模型计算该野生绢丝昆虫与2种明目大蚕蛾及其他19种柞蚕属绢丝昆虫的遗传距离,结果显示其与明目大蚕蛾的遗传距离仅为0.005;构建的进化树中,该野生绢丝昆虫也是最先与明目大蚕蛾聚在一起。此外,该野生绢丝昆虫的成虫形态特征与已有文献描述明目大蚕蛾的特征相符。综合分子鉴定结果与成虫的形态特征,确认该野生绢丝昆虫为明目大蚕蛾(Antheraea frithi)。供试明目大蚕蛾所生产的茧为黄褐色,有茧柄,无茧孔,平均茧大小为20 mm×38 mm,平均单粒茧质量6.55 g,平均茧层率10.8%,初步认为具有潜在的开发利用价值。

摘 要:
Yunnan province is one of the areas with the richest silk-spinning insect species of Saturniidae in China. In present study,molecular identification was conducted on a wild silk-spinning insect collected in Chuxiong District of Yunnan province using DNA barcoding technology. Besides,cocoon characters of this wild insect were investigated. Genomic DNA was extracted from pupae of this wild insect and amplified using universal PCR primers designed according to insect mitochondrial cytochrome oxidase subunit I gene(mt DNA COⅠ). A CO I gene fragment of 574 bp was obtained(Gen Bank accession No. KR812471). Sequence alignment of CO I gene sequence revealed a 99. 47 % sequence identity between this insect and Antheraea frithi. Kimura-2-Parameter model was taken to calculate the genetic distance of this wild insect with two varieties of Antheraea frithi and 19 other insect species of Antheraea. The genetic distance between this wild insect and Antheraea frithi Taiwan was as low as 0. 005,and in the constructed phylogenetic tree this wild insect clustered first with Antheraea frithi. Moreover,adult morphological characters of this wild insect are consistent with the recorded characters of Antheraea frithi. Based on molecular identification result and adult morphological characters,this silk-spinning insect is confirmed to be one variety of Antheraea frithi. The cocoons of this insect have brownish yellow color,have cocoon stem but have no cocoon pole. Cocoons are 20 mm wide and 38 mm long on average. Average cocoon weight is 6. 55 g,and average cocoon shell ratio is 10. 8 %,showing certain potential for commercial development and utilization.
