摘  要:
【目的】检测尾型极端差异的阿勒泰羊、湖羊和藏羊三群体中TCF7基因增强子一处SNP是否存在分化现象,从而确定该SNP是否可作为一个有效地筛选脂尾与瘦尾性状的分子标记。【方法】对三品种绵羊286份基因组进行目标SNP片段的扩增,并采用PCR-RFLP方法对以上样本扩增片段进行酶切分型,最后采用卡方检验的方法验证各基因型在各群体中的Hardy-Weinberg平衡性。【结果】高效扩增出所有样本含该SNP DNA扩增,经Mfe I酶切分型后共鉴定出了三种基因型(CC、TT和CT),其中CC基因型在瘦尾藏羊群体中的百分比高达80.3%,是脂尾型阿勒泰羊与湖羊的两倍以上。C/T等位基因的比率显示藏羊群体中这一比值分别为其他两种脂尾型绵羊品种的8倍,提示T等位基因可能与脂尾性状相关。【结论】该SNP可作为脂、瘦尾羊选育的一个分子标记。

摘  要:
【Objective】To detect the polymorphism of an mutation in TCF7 enhancer region among three sheep flocks of extremely different fat- tail types,and further to determine whether the SNP can be used as an effective molecular marker screening the trait of fat tail or thin tail.【Method】In this study,the polymerase chain reaction( PCR) was applied to amplify the DNA fragment containing the target SNP( three sheep breeds,n = 286),and PCR- RFLP method was employed to genotype,and Chi- Square test was used to verify theHardy- Weinberg equilibrium of genotype groups. 【Result 】The DNA fragments containing the SNP( the43128620 thsequence on sheep chromosome 5) were successfully amplified,and three genotypes( CC,TT and TC) were identified by Mfe I endonuclease enzyme. The frequency of CC genotype in thin- tail Tibetan sheep population was up to 80. 3%,which was nearly two times higher than that of Altay fat tail or Hu sheep. The C /T allele ratio of Tibetan sheep population was respectively 8 times higher than that of the two fat tail sheep breeds,indicating C allele may be related with the trait of the thin tail.【Conclusion】This SNP could be used as a molecular marker in fat or thin tail sheep breeding.
