两个油菜品系的fad2和sad基因序列差异比较

摘  要:
以来源相同而油酸含量差异显著的两个甘蓝型油菜DH品系WH141和WH149为研究材料克隆了与油酸含量相关的两个关键基因fad2(脂肪酸脱氢酶基因)和sad(硬脂酰ACP脱氢酶基因),并对克隆序列多次测序,利用DNAman软件分析两个基因序列差异。结果表明:①以fad2基因序列中的6个连续变异碱基为依据可将该基因分为2类,AF型和AY型。两者均存在于WH141和WH149品系中。AF型序列内两品系共有3处有义突变,但两品系对应氨基酸的出现频率有明显差异,说明相应fad2基因的拷贝数在两品系中有较大差异。另外,在氨基酸序列中第241位疏水氨基酸F变异为极性氨基酸Y,可能对该基因的功能有较大影响。AY型序列内,除了单碱基变异外,还存在小片段的插入缺失。单碱基变异大部分对应翻译不同种类的氨基酸。片段的插入缺失导致多个读码框出现,比较后发现WH141和WH149有3个相同的读码框,而且在WH149中发现还有2个可能的fad2基因特异表达框。②对比两品系sad基因的外显子序列后发现,WH141和WH149存在14个位点的单碱基变异,且绝大部分变异位点编码的氨基酸发生了改变。

摘  要:
The rapeseed DH lines WH141 and WH149,which have the same origin but different oleic acid content,were used as materials to clone the two key genes fad2(Δ12 fatty acid desaturase) and sad(stearoyl-ACP desaturase) related with oleic acid content by PCR method based on candidate gene sequences.The clones of both fad2 and sad genes were randomly selected and sequenced.Sequences were analyzed using the DNAman bio-software.The results indicated: ① Based on a six continuous bases mutation among fad2 gene sequences,the fad2 sequences were divided in two classes: AF type and AY type,which existed in two lines.The sequence comparison showed that there were 3 amino acid sensitive mutations in the AF type,but amino acids on the same loci had distinct frequencies in two lines,which meant that copy numbers of fad2 gene were different in WH141and WH149.The lyophobic amino acid F on the 241th locus in the protein mutated into hydrophilic amino acid Y,which may affect the protein function.In the AY type,besides the single base mutation,the small DNA fragments′ insertion and deletion was also found.Most of the base mutations corresponded with different classes of amino acids.The small DNA fragments′ insertion and deletion caused several new ORFs in the two lines,three of which were the same.Two new possible fad2 gene ORFs were specific in the line WH149.② The sequence comparison of the sad gene′s exons indicated that in the two lines,there existed 14 single bases mutation which encoded different classes of amino acids.
