猎豹与虎猫杯状病毒的分离及其超变区基因比较研究

摘  要:
用F8 1细胞从上海某动物园患口腔溃疡的猎豹和虎的唾液病料中分离获得两株杯状病毒 ,经形态学、理化学、生物学鉴定和病毒核酸超变区基因RT_PCR扩增与序列测定证明两株病毒均为猫杯状病毒 (FCV) ,分别命名为FCV/cheetah/Shanghai/ 0 2 / 2 0 0 2与FCV/tiger/Shanghai/ 0 3/ 2 0 0 2。人工感染猫可引起体温升高 ,口腔溃疡 ,食欲下降等症状。两株病毒的超变区序列 5 2 0bp长片段间的同源性为 99.2 % ,与国内桂林虎分离株 (TFCV9710 )的同源性为 74 .0 % ,与国外分离株的总体同源性为 5 8.1% ,说明不同宿主或同种不同个体间杯状病毒分离株超变区核酸差异十分显著 ,符合猫杯状病毒的分子生物学特征

摘  要:
Saliva of a cheetah and a tiger from Shanghai wild animals zoo,which suffered from oral ulcer,was collected and inoculated onto cultured F81 cells.Two islolates of calicivirus_like virus were obtained and identified by their physicochemical and biological characteristics.The hypervarialbe regions of virus genome were amplified by RT_PCR and analysised by sequence comparison.The results showed that the two isolates,named as FCV/cheetah/Shanghai/02/2002 and FCV/tiger/Shanghai/03/2002,respectively had specific characteristics of feline caliciviruses.Laboratory infection of cats by the virus presented a similar clinical signs of fever,and ulcer and decreased appetite.The homology of the hypervariable region between two isolates of calicivirus was 99.2 %,and 77.4 % compared with that of a tiger isolate from Guilin,China.The population homology of the two isolates with that of isolates from overseas was 58.1 %.All above showed that significant difference exited among different species or isolates,which is the most important characterization of molecular biology of feline calicivirus.
