西藏八角莲不同部位鬼臼毒素的HPLC分析

摘  要:
[目的]建立了西藏八角莲[Dysosma versipellis(Hance)M.Cheng]不同部位鬼臼毒素的HPLC分析方法,并对鬼臼毒素含量进行检测。[方法]色谱条件为:色谱柱,Whates C18柱(125.0 mm×4.6 mm,5μm);流动相,甲醇∶水(55∶45,V/V);流速,1.0 ml/min;检测波长,290 nm;柱温,25℃;进样量,20μl。线性范围为1~200μg/ml,回归方程:y=15121x-46585,r=0.9991。[结果]西藏八角莲根、块茎、叶、叶柄、花和果实中均检测到鬼臼毒素,但是不同部位含量差异较大。其中,块茎中鬼臼毒素含量最高,为(180.50±2.74)(md)μg/g,之后依次为根中(58.83±1.59)(md)μg/g,花中(40.33±1.24)(md)μg/g,叶中(36.93±1.09)(md)μg/g,叶柄中(29.90±0.45)(md)μg/g,果实中(6.03±0.12)(md)μg/g,依次降低。[结论]该检测方法灵敏度高、稳定性好、重复性高、分离效果好,不同组织中鬼臼毒素含量以块茎中最高,果实中最低。该研究可为西藏药源植物功效成分的分析提供理论与实践指导。

摘  要:
[Objective]To analyze the podophyllotoxin in different parts of D.tsayuensis(Hance)M.Cheng by HPLC,and the contents of which were determined.[Method]The chromatographic conditions were as follows: Whates C18column(125.0 mm×4.6 mm,5 μm),mobile phase of carbinol and water(55∶45),flow-rate of 1.0 ml/min,detective wavelength of 290 nm,column temperature of 25 ℃ and sample volume of 20 μl.The linear range linear range linear range was 1~200 μg/ml and the regression equation was: y=15 121x-46 585,r=0.999 1.[Result]There were podophyllotoxins in the root,tuber,leaf,petiole,flower and fruit of D.tsayuensis(Hance)M.Cheng,while the contents of podophyllotoxin varied in different parts.There was the highest level of podophyllotoxin in the tuber,(180.50±2.74)(md)μg/g;the content of podophyllotoxin in the root took the second place,(58.83±1.59)(md)μg/g;the content of podophyllotoxin in the flower was(40.33±1.24)(md)μg/g,the content of podophyllotoxin in the leaf was(36.93±1.09)(md)μg/g;the content of podophyllotoxin in the petiole was 29.90±0.45(md)μg/g;the content of podophyllotoxin in the fruit was(6.03±0.12)(md)μg/g.[Conclusion]HPLC was of high sensitivity,good stability,repeatability and separating effect;the content of podophyllotoxin was the highest in tuber and the lowest in the fruit.This research would provide theoretical and practical direction for analyzing efficiency compositions in medicinal plants of Tibet.
