NF-κB抑制剂PDTC对卵巢癌细胞HO8910增殖、凋亡的影响

摘  要:
应用四甲基偶氮唑盐比色(MTT)法检测不同浓度吡咯烷二硫代氨基甲酸(PDTC)、顺铂(DDP)和联合应用PDTC与DDP对HO8910细胞生长抑制的影响;采用流式细胞仪检测不同浓度PDTC作用HO8910后的细胞凋亡率。结果表明:1PDTC浓度为25、50、100和200μmol·L-1时,作用12、24、36h均明显抑制HO8910细胞的增殖,与对照组比较差异极显著(P<0.01),并具有时间、剂量依赖性。2顺铂联合应用PDTC时能显著提高顺铂杀伤卵巢癌细胞作用。325、50、100μmol·L-1PDTC诱导卵巢癌HO8910细胞凋亡。PDTC能有效抑制卵巢癌HO8910细胞增殖并能够增强顺铂化疗作用,提高疗效。

摘  要:
The cytotoxicity of PDTC,DDP,or PDTC combined with DDP to HO8910 cell line was detected by MTT chromatometry.The apoptotic ratios were detected by flowcytometry using PI/Annexin staining method.The results were as follows:① After exposure to 12,24,36 h,PDTC(25,50,100,200 μmol·L-1)could significantly inhibit the proliferation of HO8910 cells in a dose-dependent and time-dependent fashion(P<0.01);② Application of DDP combined with PDTC significantly improved cytotoxicity of DDP to HO8910.③ In contrast to the control,PDTC(25,50,100 μmol·L-1)could significantly induce apoptosis of human ovarian cancer HO8910 cells.It was concluded that PDTC could significantly inhibit the proliferation of HO8910 cells and enhance the effect of DDP for chemotherapy and increase the therapeutic effect.
