pH和琼脂糖浓度对大白菜不定芽和胚状体诱导的影响

摘  要:
以1039,B-17和637大白菜自交系为试材,研究了培养基中不同pH和琼脂糖质量浓度对胚状体诱导的效应。结果表明,当pH值为6.0~6.2,琼脂糖质量浓度为8~9 g/mL时,可以诱导"1039"大白菜试材产生胚状体,其诱导频率约1.0%;当pH值为5.7~5.8,琼脂糖质量浓度为8~10 g/mL时,3个大白菜试材中均有不定芽产生,并且随着琼脂糖浓度的增加,不定芽的数量增加。不同的大白菜试材间诱导频率没有明显差异,均为3.0%。

摘  要:
Through changing pH value and agarose concentration of culture medium,we developed a method for embryo and adventitious bud induction.The MS medium supplemented with 6-BA 5 mg/L,NAA 0.5 mg/L,pH 6.0~6.2,agrose 8~9 g/mL was found to be optimum for embryo induction in 1039 imbred line,and the frequency of induction was about 1.0%.pH 5.7~5.8,agrose 8~10 g/mL was found to be optimum for adventitious bud inducing in 1039,B-17 and 637 imbred lines,and with the increase of agarose concentration,frequency of induction also enhanced.The average frequency of adventitious bud induction was about 3%,indicating no significant difference among our tested lines.
