江西酸橙不同栽培变种RAPD分析

摘  要:
[目的]对江西酸橙的不同栽培变种进行RAPD分析。[方法]采用RAPD标记对江西酸橙不同栽培变种进行了种间亲缘关系和遗传多样性分析,同时建立了酸橙RAPD标记PCR扩增的最适反应条件。[结果]结果表明,酸橙RAPD-PCR扩增的最适条件为:60 ngDNA模板,2.5 UTaq酶,镁离子终浓度为2.0 mmol/L,dNTPs终浓度为150μmol/L,PCR反应总体积为25μl;酸橙不同栽培变种之间存在一定的遗传距离,但是同一品种各单株之间遗传差异不大。[结论]为江西酸橙良种培育提供基础的理论依据。

摘  要:
[Objective] The research aimed to make the random amplified polymorphic DNA(RAPD) analysis of different cultivars of Jiangxi Citrus aurantium Linn.[Method] RAPD method was used to study the affinity and genetic diversity of different cultivars of Citrus aurantium from Jiangxi Province,and at the same time,the best reaction conditions of RAPD-PCR amplification were established.[Result] The results showed that the most optimal reaction system of RAPD-PCR was as follow: 60 ng template DNA,2.5 U Taq polymerase,Mg2+ concentration 2.0 mmol/L,dNTPs concentration 150 μmol/L in the 25 μl reaction volume.It was discovered that a certain genetic distance existed in different cultivars.However,genetic differences in individuals of the same variety were not distinct.[Conclusion] The study can provide the theoretical basis for breeding the good variety of Citrus aurantium of Jiangxi Province.
