ESR1与PGR基因在不同繁殖力小尾寒羊下丘脑-垂体-卵巢轴的表达研究

摘  要:
本试验旨在揭示ESR1和PGR基因在小尾寒羊多羔性状中的作用,深入研究绵羊多羔性状的机理。采用半定量反转录-聚合酶链式反应结合实时荧光定量PCR技术检测ESR1和PGR基因在不同繁殖力小尾寒羊群体12个组织(小脑、下丘脑、垂体、卵巢、输卵管、子宫、心脏、肝脏、脾脏、肺脏、肾脏)中的表达差异。结果表明:ESR1和PGR基因在不同繁殖力小尾寒羊群体的12个组织中均有表达;ESR1和PGR基因分别在小尾寒羊子宫和输卵管中高表达;多羔小尾寒羊子宫、输卵管、卵巢和下丘脑组织中ESR1基因的表达量与单羔小尾寒羊无显著差异(P>0.05);多羔小尾寒羊输卵管、子宫、卵巢和下丘脑组织中PGR基因的表达量极显著高于单羔小尾寒羊群体(P<0.01)。结果提示,PGR基因可能参与小尾寒羊多羔性状的调控。

摘  要:
To reveal the roles of ESR1 and PGR genes in the high fecundity of sheep and research the mechanism of prolificacy in sheep. Semi-quantitative reverse transcriptase-polymerase chain reaction and real-time fluorescence quantitative PCR were performed to detect ESR1 and PGR mRNA expression in twelve tissues of multiparous and uniparous Small Tail Han sheep. The results showed that both ESR1 and PGR genes were expressed in twelve tissues of Small Tail Han sheep. The expression of ESR1 and PGR was high level in uterus and oviduct of multiparous and uniparous Small Tail Han sheep, respectively. The expression of ESR1 in uterus, oviduct, ovary of multiparous STH was no significantly different than that in uniparous STH(P>0.05). The expression of PGR in hypothalamus, pituitary, uterus, ovary and oviduct in multiparous STH was significantly higher than that in uniparous STH(P<0.01). The results suggested that PGR gene may be involved in the regulation of multiple birth in sheep.
