草分枝杆菌胞壁肽聚糖的分离提取和鉴定

摘  要:
本试验以草分枝杆菌为研究材料,扩大培养后采用超声波破碎从草分枝杆菌中分离提取得到大量细胞壁,然后经三氯乙酸、乙醚、胰蛋白酶缓冲液处理等操作,分离得到较纯净肽聚糖并对其组成成分和结构展开了分析.通过称重计算提取肽聚糖得率为13.33%.溶菌酶溶解试验9 h后眼观变为澄清.红外光谱(IR)分析,提取的肽聚糖样本与肽聚糖已知结构特征相一致.经过聚丙烯酰胺凝胶电泳(SDS-PAGE)分析,经溶菌酶水解后的肽聚糖分子量约为62 ku.

摘  要:
In this experiment, the Mycobacterium phlei was used as research materials, after expanding training, ultrasonic broken was optimized so as to obtain a large number of cell walls.And then by trichloroacetic acid, ethyl ether, trypsin buffer processing operations, purified peptidoglycan was isolated. Through weighing the peptidoglycan extracted, the yield ratio was13.35%. In lysozyme experiment, 9 h later, the peptidoglycan became clarifying. With infrared spectroscopy(IR) analysis, the extracted peptidoglycan had the same characteristics with samples we knew. With Gel electrophoresis(SDS-PAGE) analysis, the molecular weight of peptidoglycan was about 60 ku.
