水稻种子胚乳蛋白的双向电泳分析技术

摘  要:
为探讨水稻灌浆期间与籽粒充实相关蛋白表达的变化,建立了一套适于水稻种子胚乳蛋白双向电泳分析技术,探讨了胚乳蛋白的提取、纯化以及双向电泳的一些关键技术.结果表明,通过乙醇铵沉淀、高压等电聚焦和两步平衡等方法可以获得较好的分析结果,获得胚乳蛋白斑点数约 350 个,等电点范围在 3.0~9.0,其中大部分集中在 4.0~7.0.

摘  要:
In order to research relative proteins with grain plumpness during filling stage of rice,one serial of 2-D electrophoresis which be suitable to endosperm proteins of rice seed was set up.It was discussed on the extraction, purification of the endosperm proteins and some key technologies of 2-D electrophoresis.The results were showed that, by precipitating with ammonium ethanol isoelectric focusing under high-voltage and two-step equilibrium,about 350 protein spots that the isoelectric point ranges from 3.0 to 9.0 were found,and the major isoelectric point concentrated on 4.0 to 7.0.
