分子技术在蜜蜂白垩病病原鉴定和诊断上的应用

摘  要:
蜜蜂白垩病是一种由球囊菌属(Ascosphaera)的真菌寄生而引起蜜蜂幼虫死亡的传染性疾病,对蜂群的群势发展影响极大,近几年又呈现出大面积爆发的趋势。但目前对于该病的诊断和病原鉴定多依赖于典型症状,方法极不可靠。本文介绍了可用于分子鉴定和诊断的的研究基础,探讨利用球囊菌属的各病原rDNA的ITS1-5.8S-ITS2序列资料,分别设计出高特异性引物,用PCR技术扩增出可用于鉴定病原物种的分子标记,从而构建一套快速、准确诊断该病的分子技术。

摘  要:
Fungi in the genus Ascosphaera are the causative agents of chalkbrood, a major infectious disease affecting bee larval viability and causing serious collapse of entire colonies. The trend of the disease extensive outbreak has emerged in China in the past few years. However, most diagnostic methods of chalkbrood based on typical clinical symptoms have bee not reliable. The achievements in researches on the molecular level for identification and diagnosis of Ascosphaera are reviewed in this paper. And the feasibility of develop a quick and reliable diagnostic method for identifying Ascosphaera infections in honey bees using molecular markers amplified by PCR designing the highly specific primers based on the DNA sequences data of the 5.8s ribosomal DNA and the internal transcribed spacers (ITS1 and ITS2) from species of Ascosphaera is discussed.
