CiF3H基因克隆及功能分析

摘  要:
黄烷酮-3-羟化酶(F3H)是类黄酮合成途径的中枢位点,调控代谢途径中黄酮醇和花青素合成。文章根据转录组数据库中F3H基因中间片段设计特异引物,以中间锦鸡儿cDNA为模板,克隆得到CiF3H基因全长,通过序列分析、系统进化分析加以确认。qRT-PCR检测分析发现,中间锦鸡儿CiF3H基因受紫外、NaCl和ABA等胁迫诱导时表达模式具有相同规律。CiF3H基因在叶和根中表达量相当,茎中较少。异源表达CiF3H基因拟南芥的总黄酮含量少量增加,花青素含量降低。

摘  要:
Flavanone-3-hydroxyase(F3 H) is the essential site in flavonoids biosynthesis pathway,which regulates the synthesis of flavonols and anthocyanins in the whole metabolic pathway. In this study, specific primers were designed according to the intermediate fragment of F3 H gene in the transcriptome database, and the full length of CiF3 H gene sequence was cloned using Caragana intermedia cDNA as template, which was confirmed by sequence and phylogenetic analyses. The expression of CiF3 H had the same pattern by ultraviolet(UV), NaCl and abscisic acid(ABA) stresses as revealed by quantitative real-time PCR(qRT-PCR). The expression of CiF3 H was similar in leaves and roots, but less in stems. The content of total flavonoids in transgenic Arabidopsis increased slightly, but the content of anthocyanins decreased.
