绵羊胚胎不同发育阶段及其组织中MicroRNA-483-5p表达分析

摘  要:
【目的】研究MicroRNA(miR)483-5p在中国美利奴羊胚胎不同发育阶段肾脏和脐带组织中的表达变化,以了解其在绵羊胚胎发育中的重要作用。【方法】选择中国美利奴羊胚胎不同发育时期(45、85、115和135 d 4个时期)肾脏和脐带组织为研究材料,采用半定量RT-PCR检测第45 d绵羊胚胎不同组织中miR-483-5p的表达;同时利用实时荧光定量PCR对miR-483-5p在绵羊胚胎不同发育阶段肾脏和脐带组织的表达变化进行实时检测。【结果】miR-483-5p在所检测的绵羊45 d胚胎各组织中广泛表达,其中以肌肉、心脏和肺脏组织表达丰度最高。实时荧光定量PCR结果显示,不同发育阶段绵羊胚胎肾脏和脐带组织miR-483-5p的表达具有明显的时空选择性。脐带组织miR-483-5p随着胚胎发育其表达量呈逐渐下降的趋势;肾脏组织miR-483-5p则是随着胚胎发育在第85 d其表达量下降到最低,其后在115和135 d其表达量又逐渐上升。不同发育阶段胚胎肾脏、脐带组织miR-483-5p的表达量呈显著性差异(P<0.05)。【结论】miR-483-5p在绵羊胚胎不同发育阶段肾脏或脐带组织中表达明显不同,表明其表达具有选择性;依据miR-483-5p表达的差异性及其涉及的生物学功能,推测miR-483-5p在绵羊脐带和肾脏组织中具有一定的调控作用。研究为进一步了解miR-483-5p在胚胎肾脏和脐带发育中的作用机制提供一定的理论依据。

摘  要:
【Objective】The aim of this study is to detect the expression of MicroRNA( miR) 483-5p in kidney and umbilical cord tissues at different stages of embryos in Chinese Merino and to explore the roles of miR-483-5p in embryonic development. 【Method】In this study,different development periods( 45 d,85 d,115 d and 135 d 4 times) kidney and umbilical cord tissue of Chinese Merino sheep embryo were taken as the research material,the expression of miR-483-5p in different tissues of 45 th day embryos of sheep was analyzed by RT-PCR. On the basis,the expression of miR-483-5p in kidney and umbilical cord tissues at different stages of embryos were analyzed and compared by Real-time PCR.【Result】The results showed that miR-483-5p were widely expressed in the tested tissues,in which the expression level of muscle,heart and lung was much higher than the others. The real-time PCR showed that the expression of miR-483-5p in kidney and umbilical cord tissues at different stages embryos of sheep had obvious temporal selectivity. In the wake of embryo development the expression of miR-483-5p in umbilical cord tissues was gradually decreased,but in kidney tissues,the expression of miRNA 483-5p was declined to the lowest in 85 d of embryonic development,then gradually increased in the 115 d and 135 d. There were significant differences for expression change extent of miR-483-5p in kidney or umbilical cord in different development stages of embryo.【Conclusion】These results suggested that the expression of miR-483-5p in kidney or umbilical cord at different development stages and various tissues of embryos were obviously different,which suggested that miRNA 483-5p was selectively expressed in sheep embryonic development. It was deduced that miR-483-5p had regulation function in kidney and umbilical cord according to the difference of expression and the reported function of miR-483-5p. These results might provide the theoretical basis for further study of mechanism of microRNAs in kidney and umbilical cord of sheep embryo.
