家兔BMPR1B基因多态性及其与生长速度的相关性分析

摘  要:
本文旨在研究家兔BMPR1B基因的多态性及其与生长速度的相关性。采用PCR产物测序方法检测BMPR1B基因编码区的多态性,利用PCR-RFLP技术对候选SNPs位点(c.117G>A)进行基因分型,研究其与家兔生长速度的关联性(N=262)。结果表明,在BMPR1B基因编码区共存在14个SNPs位点,它们分别是c.9C>T、c.54C>T、c.75T>A、c.88C>T、c.96T>C、c.117G>A、c.384A>G、c.392C>T、c.540C>T、c.603T>C、c.1254T>A、c.1332A>G、c.1356A>G及c.1436A>G,除c.392C>T突变致使该基因编码的第131个氨基酸亮氨酸(leu)被丝氨酸(ser)替换外,其他13个位点均为同义突变。c.117G>A位点处于Hardy-Weinberg平衡状态(P>0.05),为中度多态(0.25A位点AA基因型35日龄断奶重显著高于GG基因型和AG基因型(669.556±43.733VS 612.571±10.588VS 612.478±13.73)。

摘  要:
In this paper,the aim of this experiment was to study the association between the polymorphisms of BMPR1 B gene and body weight in rabbits. The polymorphisms of BMPR1 B gene were detected by direct sequencing,the SNPs were genotyped by PCR-RFLP method,and the association between c. 117 G > A and body weight in 262 young New Zealand rabbis at different age stages was studied. The result showed that a total of 14 coding SNPs were found in BMPR1 B gene. Among them,c. 9C > T,c. 54 C > T,c. 75 T > A,c. 88 C > T,c. 96 T > C,c.117 G > A,c. 384 A > G,c. 540 C > T,c. 603 T > C,c. 1254 T > A,c. 1332 A > G,c. 1356 A > G,and c. 1436 A > G were synonymous SNPs,but c. 392 C > T was non-synonymous SNP which led leucine to serine. SNP of c. 117 G > A was at Hardy-Weinberg equilibrium( P >0. 05) and presented moderate polymorphism( 0. 25 < PIC < 0. 50). The association analysis indicated that weight of BMPR1 B gene AA genotype individuals weaning BW35 at c. 117 G > A site was significantly greater than that of GG and GA genotype individuals( P < 0. 05)( 669.556 ±43.733 VS 612.571 ±10.588 VS 612.478 ±13.73).
