脂肪细胞膜蛋白卵黄抗体对脂肪细胞增殖与凋亡的影响

摘  要:
以0.1、1.0和10.0μg/mL 3种剂量脂肪细胞膜蛋白卵黄抗体处理体外培养的3T3-L1前脂肪细胞或经其诱导分化的脂肪细胞,采用四唑盐(MTT)比色法检测3T3-L1前脂肪细胞增殖能力,以乳酸脱氢酶漏出率为指标观察脂肪细胞膜蛋白卵黄抗体对脂肪细胞的细胞毒性,用流式细胞仪、Hoechst 33258荧光染色分析脂肪细胞凋亡情况以及油红O染色法测定脂肪细胞脂肪含量。结果表明:用不同质量浓度(0.1、1.0和10.0μg/mL)的脂肪细胞膜蛋白卵黄抗体处理可抑制3T3-L1前脂肪细胞增殖,并影响脂肪细胞的脂肪合成(P<0.05),但对脂肪细胞无明显的细胞毒作用;还可见凋亡峰和典型的凋亡形态学改变,流式细胞仪检测出的脂肪细胞凋亡率分别为1.62%、2.33%和4.57%,呈浓度依赖性,与对照组(0.36%)比较,差异皆显著(P<0.01)。提示:脂肪细胞膜蛋白卵黄抗体可抑制前脂肪细胞增殖,诱导脂肪细胞凋亡并影响脂肪合成。

摘  要:
The 3T3-L1 preadipocytes or differentiated adipocytes were treated with 0.1,1.0,10.0 μg/mL yolk antibody against adipocyte plasma membranes(AIgY) for 12,24,48 or 72 hours respectively.The proliferation activity of 3T3-L1 preadipocytes was examined by MTT spectrophotometry.The degree of adipogenesis was measured by Oil Red O staining extraction assay.Complement-mediated cytotoxicity of adipocytes was determined through leakage rate of LDH(lactate dehydrogenase),while apoptosis was analyzed by flow cytometry and Hoechst 33258 fluorescence staining.The proliferation activity of 3T3-L1 preadipocytes and adipogenesis of adipocytes were substantially decreased by the treatment with AIgY but complement-mediated cytotoxicity was not observed.Cytoplasmic fat content was reduced significantly.Apoptotic peak and typical morphological changes in apoptosis could be observed in cells exposed to 0.1,1.0,10.0 μg/mL yolk antibody against adipocyte plasma membranes for 48 h,and apoptotic rate determined by flow cytometry was significantly higher than that in control groups in a dose-related manner(P<0.01).These results showed that the yolk antibody against adipocyte plasma membranes could inhibit the proliferation of 3T3-L1 preadipocytes,affect adipogenesis of adipocyes and induce the apoptosis of adipocytes.The apoptotic indexes were dependent on the concentration.
