水貂IGF-1R基因SNPs筛查及其与生长性状的关联分析

摘  要:
为分析IGF-1R基因部分SNPs与银蓝水貂与美国短毛黑水貂2个群体生长性状的关联性。以同龄同场的银蓝水貂和美国短毛黑水貂为研究对象,采用PCR产物直接测序法筛查IGF-1R基因CDS序列的多态位点,利用SPSS(19.0)软件GLM统计模型分析基因SNPs不同基因型与水貂生长性状的相关性。在线软件对IGF-1R基因进行生物信息学分析。结果表明:在IGF-1R基因外显子2和外显子8分别检测到G207A同义突变和G1782A同义突变,根据测序峰图对2位点进行基因分型。群体遗传学分析显示,G207A位点在银蓝水貂群体中属于低度多态,G1782A位点在银蓝水貂群体中属于中度多态,G207A和G1782A 2个位点在美国短毛黑水貂中均为中度多态;χ~2检验表明,银蓝水貂和美国短毛黑水貂群体在2个突变位点处于HardyWeinberg平衡状态(P>0.05);关联分析表明,G1782A位点与美国短毛黑水貂的体重显著相关(P<0.05),突变纯合子AA基因型个体的体重值显著高于GG基因型个体和GA基因型个体。因此G1782A位点适合于短毛黑水貂体重的分子标记选择。

摘  要:
The aim of this study was to analyze the correlation of IGF-1R gene polymorphisms with some economic traits in mink. Sliver blue mink and American mink of the same age and in the same field were chosen as subjects. The technology of direct sequencing of PCR products was used to detect single nucleotide polymorphismof IGF-1R gene in CDS,and its correlation of polymorphisms and different genotypes and combined genotypes with growth traits was analyzed by SPSS( 19. 0). Meanwhile,bioinformatics analysis of IGF-1R gene was conducted with online software. Two SNPs( G207 A,G1782A) of IGF-1R gene were indentified in the population,which were exon2 and exon8.Through population genetic analysis,the results showed that G207 A loci was at low polymorphic status; G207 A and G1782 A loci were at moderate polymorphic status. The χ~2 tests showed that the 4 loci were in the status of Hardy-Weinberg equilibrium( P>0. 05) in sliver blue and American mink.The gene-specific SNP marker correlation analysis was investigated. Marbling score was significantly higher in individual with AA genotype than in individual with AG and GG genotypes of G1782A( P<0. 05). The results suggest that G1782 A may be the useful marker in weight of the American mink.
