摘  要:
为建立黄蜀葵花中主要活性成分金丝桃苷及槲皮素含量的高效液相色谱(HPLC)测定方法,以金丝桃苷和槲皮素标准品为对照,色谱柱采用COSMOSIL5C18-AR-Ⅱ(4.6mm×250mm,5μm),流动相为0.4%磷酸溶液、乙腈,柱温为35℃,波长为360nm,流速为1.0mL·min-1,梯度洗脱分离。结果表明:金丝桃苷回归方程为y=351.5x+202.1(r=0.999 5),金丝桃苷浓度为5.296~52.96μg·mL-1时呈良好的线性关系;槲皮素回归方程为y=364.9x+23.84(r=0.999 6),槲皮素浓度为5.272~52.72μg·mL-1时呈良好的线性关系。该方法操作简便、重复性好、精密度高,可作为黄蜀葵花中金丝桃苷和槲皮素的质量控制方法。

摘  要:
A method based on HPLC was used to determine the content of hyperfine and quercetin in Abelmoschi corolla.With hyperfine and quercetin as refence substances,the separation was performed on COSMOSIL5C18-AR-Ⅱ(4.6mm×250mm,5μm)column with the mobile phase consisted of 0.4% phosphoric acid-acetonitrile.The column temperature was 35 ℃ and detection wavelength was set at 360 nm,at the flow rate of 1.0mL·min-1 with a nonlinear gradient elution program.The test showed that within the concentration range 5.296-52.96μg·mL-1,the linear correlation equation of hyperfine was y=351.5x+202.1,with a coefficient of determination r=0.999 5and that within the concentration range 5.272-52.72μg·mL-1,the linear correlation equation of quercetin was y=364.9x+23.84,with a coefficient of determination r=0.999 6.The method was simple,rapid and accurate and can be adopted for the quality control of A.corolla.
