丙磺舒对阿莫西林在猪体内的药代动力学影响

摘  要：
本实验采用高效液相色谱法测定猪血浆中阿莫西林的浓度,研究了丙磺舒对阿莫西林在健康猪体内药物动力学的影响。结果表明,单剂量肌肉注射阿莫西林后,阿莫西林吸收迅速,药峰时间为(0.19±0.11)h,药物浓度为(7.21±3.27)μg/mL,其动力学模型为一级速率一室模型。使用丙磺舒后,阿莫西林的半衰期由(0.86±0.18)h延长到(2.77±1.02)h(P<0.05),AUC由(12.38±5.69)μg/mL·h增加至(35.24±18.62)μg/mL·h,药峰时间为(0.42±0.32)h,药峰浓度为(6.24±3.43)μg/mL·h。

摘  要：
Five healthy pigs were used in a randomized design.In repeated experiments all pigs were administered of 10 mg of probenecid per kg body weight by intramuscularly injection and 7 days later they were administered of 10 mg of probenecid per kg body weight by intramuscular injection,followed immediately by 10 mg amoxicillin per kg body weight by intramuscular injection.A rapid and sensitive assay for amoxicillin in pgi plasma was developed using reversed phase high performance liquid chromatography (HPLC).The result was as follows:The analytical recovery of amoxicillin from plasma was >82.5%,and the relative standard deviations for withinday and betweenday were<5.00% and <10.98% for treatment with amoxicillin alone, the elimination half time(T1/2)was (0.86±0.18) h,AUC was (12.38±5.69) μg/mL·h,the peak levels of amoxocillin in plasma averaged (7.21±3.27) μg/mL at (0.19±0.11) h. And preceding treatment with pr obenecid resulted in a increase in both T1/2(2.77±1.02) h(P<0.05)and AUC(35.24±18.62) μg/mL·h.
