山西省杜鹃花属植物亲缘关系的RAPD分析

摘  要:
采用RAPD分子标记技术,对山西省2种野生杜鹃和6个省内常见园艺品种进行亲缘关系分析。从50条引物中筛选出4条多态性丰富的引物,对8种杜鹃花共11个样本的DNA进行PCR扩增,其产物用琼脂糖凝胶电泳进行检测。结果表明,共扩增出44条谱带,其中,多态性谱带28条,多态位点率63.64%。用NTSYS软件计算出遗传相似系数(GS)在0.238 8~0.970 2,平均值为0.540 6,说明各样本间遗传基础差异较大;当GS为0.78时,可将供试样本分为3大类群,第1类为照山白,第2类为迎红杜鹃、羊踯躅、刺毛杜鹃和马银花,第3类为满山红、锦绣杜鹃和皋月杜鹃;第2,3类群的聚类结果表现出样品在形态学上虽有差异,但在基因水平上却有很高的同源性。RAPD标记技术从分子水平展示了杜鹃花属8种植物的亲缘关系,可为今后山西省野生杜鹃的育种和开发提供依据。

摘  要:
By using RAPD molecular markers technique, the genetic relationship between 2 wild Rhododendron species and 6 common horticultural varieties in Shanxi province was analyzed. 4 primers with rich polymorphisms were selected from 50 primers, and the DNA of 11 samples from 8 Rhododendron species were amplified by PCR. Their products were electrophorized by agarose gel electrophoresis. The result showed that a total of 44 bands were amplified, including 28 polymorphic bands and the polymorphic loci rate was 63.64%. The genetic similarity coefficient(GS)calculated by NTSYS software was 0.238 8-0.970 2, average value was 0.540 6,indicating that the genetic basis among the samples varied greatly. When the GS value was 0.78, the selected samples could be divided into three groups, the first group was Rhododendron micranthum, the second included Rhododendron mucronulatum, Rhododendron molle, Rhododendron championae and Rhododendron ovatum, the third included Rhododendron mariesii, Rhododendron pulchrum and Rhododendron indicum. The clustering results of the second and third groups showed that the samples were different in morphology, but had highly genetic homology. The RAPD marker technique reveals the genetic relationship of 8 Rhododendron species at molecular level,which will provide the basis for breeding and development of wild Rhododendron in Shanxi province in the future.
